不同干燥方法对山豆根主要有效成分含量的影响 被引量:8

Effect of Different Drying Methods on the Active Components of Sophorae Tonkinensis Radix et Rhizoma

摘要 目的:比较不同干燥方法对山豆根有效成分含量的影响,为山豆根的产地初加工提供科学依据。方法:分别采用晒干、冷冻干燥、不同温度(60,80,100℃)烘干加工处理山豆根,采用高效液相色谱法测定山豆根中主要功效成分氧化苦参碱、苦参碱、金雀花碱、三叶豆紫檀苷和芒丙花素的含量。以Waters Sunfire ODS C18柱(250 mm×4.6 mm,5μm)为色谱柱,流动相为乙腈-0.01 mol·L^-1醋酸铵(氨水调节p H至8.0),流速为0.8 ml·min^-1,检测波长为210 nm(金雀花碱、氧化苦参碱、苦参碱)、280 nm(三叶豆紫檀苷、芒丙花素),柱温为30℃。结果:综合评价结果表明,不同干燥方法下有效成分含量由高至低的排序为:80℃烘干>100℃烘干>60℃烘干>冷冻干燥>自然晒干。结论:80℃干燥方式为5种干燥方式中最优的山豆根干燥方法,可为山豆根的产地加工提供借鉴。 Objective:To provide a scientific basis for the original processing of Sophorae tonkinensis radix et rhizoma by comparing the effect of different drying methods on the contents of active components.Methods:Sophorae tonkinensis radix et rhizome was treated with different drying methods including natural drying in the sun,freeze-drying,and drying at different temperatures(60,80 and 100℃).The analysis were carried out on a Waters Sunfire ODS C18(250 mm×4.6 mm,5μm)column with the mobile phase consisting of acetonitrile-0.01 mol·L^-1 ammonium acetate solution(p H 8.0)in a gradient mode at a flow rate of 1.0 ml·min^-1.The detection wavelength was set at 210 nm for cytisine,oxymatrine and matrine,and 280 nm for trifolirhizin and formononetin.The column temperature was set at 30℃.Results:The results of comprehensive evaluation showed that the content order of active compositions after different drying methods was as follows:drying at 80℃>drying at 100℃>drying at 60℃>freeze-drying>natural drying in the sun.Conclusion:The best drying method for Sophorae tonkinensis radix et rhizoma is drying at 80℃,which can provide reference for the drying processing of Sophorae tonkinensis radix et rhizoma.
