摘要
结直肠癌是最常见的恶性肿瘤之一,在我国发病率呈逐渐上升趋势。基于分子标志物的靶向药物治疗使得晚期结直肠癌患者整体预后获得显著改善,中位生存期达到30个月以上。此外,分子标志物在结直肠癌的诊断、分期和预后预测方面也显示出重要的作用。本文对近年来有关鼠肉瘤病毒癌基因(Kirsten rat sarcoma viral oncogene homolog,KRAS)、V-raf鼠肉瘤病毒癌基因同源物B1(V-raf murine sarcoma viral oncogene homolog B1,BRAF)、磷脂酰肌醇-3-激酶催化亚单位α基因(phosphatidylinositol-3 kinasecatalytic alpha,PIK3CA)、人类表皮生长因子受体2(human epidermal growth factor receptor 2,HER-2)、人第10号染色体缺失的磷酸酶及张力蛋白同源的基因(phosphatase and tensin homologue deleted on chromosome 10,PTEN)、血管内皮生长因子(vascular endothelial growth factor,VEGF)和血管内皮生长因子受体(VEGF receptor,VEGFR)等常见的分子靶点在结直肠癌中的研究做一综述,旨在为结直肠癌个体化治疗提供参考。
Colorectal cancer is one of the most common malignancies,and its incidence is on the rise in China.Targeted drug therapy based on molecular markers significantly improved the overall prognosis of patients with advanced colorectal cancer,with a median survival of more than 30 months.In addition,molecular markers also play an important role in the diagnosis,staging and prognosis prediction of colorectal cancer.Studies on common molecular targets such as rat sarcoma viral oncogene homolog(KRAS),V-raf sarcoma viral oncogene homolog B1(BRAF),phosphatidylinositol-3 kinasecatalytic alpha(PIK3CA),human epidermal growth factor receptor 2(HER-2),phosphates and tensin homologue genes deleted on chromosome 10(PTEN),vascular endothelial growth factor(VEGF)and VEGF receptor(VEGFR)were reviewed in colorectal cancer,in order to provide references for individualized treatment of colorectal cancer.
