摘要
在单因素实验的基础上,采用正交实验对绞股蓝总黄酮提取工艺进行优化,并通过测定其对羟基自由基和超氧阴离子自由基的清除率来评价其抗氧化活性。结果表明,在提取时间为150 min、提取温度为90℃、乙醇体积分数为40%、固液比为1∶70(g∶mL)、pH值为8、虹吸次数为24次的最优工艺条件下,绞股蓝总黄酮提取量可达83.295 mg·g^-1;当绞股蓝总黄酮浓度为0.18 mg·mL^-1时,其对羟基自由基的清除率达到56.757%;当绞股蓝总黄酮浓度为0.20 mg·mL^-1时,其对超氧阴离子自由基的清除率达到65.151%。
On the basis of single-factor experiments,we optimized the extraction process of total flavonoids from Gynostemma pentaphyllum by orthogonal experiments,and evaluated their antioxidant activity by measuring scavenging rate of hydroxyl free radical and superoxide anion free radical.The results show that the content of total flavonoids from Gynostemma pentaphyllum reaches 83.295 mg·g^-1 under the optimum conditions as follows:the extraction time of 150 min,the extraction temperature of 90℃,the ethanol volume fraction of 40%,the solid-liquid ratio of 1∶70(g∶mL),the pH value of 8,and the extraction times of 24.When the concentration of total flavonoids from Gynostemma pentaphyllum is 0.18 mg·mL^-1,the scavenging rate of hydroxyl free radical is 56.757%.When the concentration of total flavonoids from Gynostemma pentaphyllum is 0.20 mg·mL^-1,the scavenging rate of superoxide anion free radical is 65.151%.
