摘要
为筛选出引起许昌烟草根腐病的病原菌,以具有典型根腐病症状的烟草病株及其根际土壤作为材料,采用组织块分离法、梯度稀释法和划线分离法筛选病原菌,通过菌株形态特征及rDNA-ITS分子生物学方法对病原菌进行鉴定,病原菌对烟草的致病性采用灌根接种法进行验证。结果表明,从烟草病株和根际土壤中筛选出38株疑似病原菌,其中菌株YG-1、W2G-2、XG-1、YJ-6、XJ-2、W1J-22与尖孢镰刀菌Fusarium oxysporum的相似性达到99.52%以上,而且对烟草幼苗具有致病性。因此,将引起许昌烟草根腐病的病原菌鉴定为尖孢镰刀菌,为烟草根腐病诊断、防控与防治提供一定的指导意义。
In order to screen out pathogen species causing tobacco root rot disease in Xuchang,tobacco plants with typical black root rot and their rhizosphere soil were sampled to isolate pathogens by organization separation method and gradient dilution method culture.The species of pathogens were identified by observing morphology characteristics and analyzing sequences of rDNA-ITS.Their pathogenicities were detected by irrigating the roots of tobacco seedlings.The results showed that 38 strains,among them,YG-1,W2G-2,XG-1,YJ-6,XJ-2,W1J-22 had more than 99.52%similarity with Fusarium oxysporum,and they were pathogenic to tobacco seedlings.Therefore,the identification of Fusarium oxysporum as the pathogen of root rot in Xuchang can provide theoretical basis for the diagnosis and control of tobacco root rot.
