摘要
目的探讨内质网应激相关蛋白和宫颈癌细胞系中人乳头瘤病毒(human papilloma virus HPV)感染的相关性。方法选择2017年1月-2018年6月在河南大学淮河医院妇产科接受治疗的宫颈癌患者60例为研究对象,对患者的临床资料进行分析,利用免疫组化法对不同HPV亚型(高危型HPV-16、HPV-18,低危型HPV-6和HPV阴性)患者的葡萄糖蛋白78(glucose regulated protein 78, GRP78)、C-Jun氨基末端激酶(C-Jun NH2-terminal kinase, JNK)、增强子结合蛋白同源蛋白(enhancer binding protein homologous protein, CHOP)3种内质网应激相关蛋白的表达水平进行分析。结果不同亚型HPV感染宫颈癌患者病理分化、病理分期、淋巴转移比较差异有统计学意义(P<0.05)。高危型HPV-16、HPV-18患者的宫颈癌病理分化以低分化为主,Ⅲ、Ⅳ期临床分期患者比例高于Ⅰ、Ⅱ期患者,患者淋巴转移率较高。高危型HPV-16、HPV-18宫颈癌患者内质网应激相关蛋白(葡萄糖蛋白78、C-Jun氨基末端激酶及增强子结合蛋白同源蛋白)的阳性表达倾向(强阳性率和阳性率)明显高于低危型HPV-6和HPV阴性宫颈癌患者(P<0.05)。结论高危型HPV-16、HPV-18宫颈癌患者的内质网应激相关蛋白表达率明显高于低危型HPV-6和HPV阴性患者,宫颈癌细胞系中人乳头瘤病毒感染的发生或与内质网应激相关蛋白表达水平有关。
OBJECTIVE To explore the correlation between the endoplasmic reticulum stress-related protein and human papillomavirus infection in cervical cancer cell lines. METHODS A total of 60 patients with cervical cancer who were treated in obstetrics and gynecology department of Huaihe Hospital, Henan University from Jan 2017 to Jun 2018 were recruited as the study objects, the clinical data of the patients were analyzed.The levels of the three types of endoplasmic reticulum stress-related proteins of the patients with different HPV subtypes(high-risk HPV-16, HPV-18,low-risk HPV-6 and HPV-negative), including glucose regulated protein 78(GRP78), C-Jun NH2-terminal kinase(JNK) and enhancer binding protein homologous protein(CHOP), were observed by means of immunohistochemical method. RESULTS There were significant differences in the pathological differentiation, pathological stage and lymphatic metastasis among the cervical cancer patients with different subtypes of HPV infection(P<0.05).The patients with low differentiation were dominant among the cervical cancer patients with high-risk HPV-16 and HPV-18, the percentage of the patients with clinical stage Ⅲ and Ⅳ was higher than that of the patients with stage Ⅰ and Ⅱ, and the rate of lymphatic metastasis was high.The positive expression tendencies(strong positive rate and positive rate) of endoplasmic reticulum stress-related proteins GRP78, JNK and CHOP of the patients with high-risk HPV-16 and HPV-18 were significantly higher than those of the patients with low-risk HPV-6 and the HPV-negative patients(P<0.05). CONCLUSION The rates of expression of the endoplasmic reticulum stress-related proteins of the patients with high-risk HPV-16 and HPV-18 are significantly higher than those of the patients with low-risk HPV-6 and HPV-negative patients.The incidence of HPV infection in the cervical cancer patients is associated with the levels of the endoplasmic reticulum stress-related proteins.
