摘要
目的研究microRNA x凋亡信号通路Y在人巨细胞病毒(HCMV)感染致神经细胞凋亡中的机制。方法选取2015年1月-2016年12月60例HCMC感染致神经细胞凋亡患者为观察组,选择正常凋亡60例为对照组,选择60例体检正常者为健康组,分析miRNAs在健康人、正常神经凋亡及HCMC感染致神经细胞凋亡患者中的表达。结果健康组、对照组miR-27a、miR-664b表达低于观察组,miR-30a表达高于观察组(P<0.05);TNM分级Ⅲ级~Ⅳ级、肿瘤数量>1个、肿瘤>5 cm及有血管侵犯HCMC感染致神经细胞凋亡血清miR-27a高表达率高于TNM分级为Ⅰ级~Ⅱ级、肿瘤数量1个、肿瘤≤5 cm及无血管侵犯HCMC感染致神经细胞凋亡患者(P<0.05);转染48 h、72 h后,HCMC感染致神经细胞凋亡细胞Caspase-3增殖计数高于未转染组(P<0.05)。结论microRNA x控制HCMV病毒复制,在HCMV感染致神经细胞凋亡中有抗病毒作用,可为HCMV感染治疗靶向点。
Objective To study the role and mechanism of microRNA x/apoptotic signaling pathway Y in neuronal apoptosis induced by HCMV infection.Methods A total of 60 patients with neuronal apoptosis caused by primary HCMC infection admitted to our hospital from January 2015 to December 2016 were selected as observation group.Another 60 patients with normal neuronal apoptosis were selected as control group.During the period 60 healthy people were selected as the the healthy group,so as,to analyze the differential expression of miRNAs in healthy people,normal neuronal abnormal patients and patients with neuronal apoptosis induced by HCMC infection.Results The expressions of miR-27a and miR-664b in the healthy group and the control group were lower than those in the observation group,and the expressions of miR-30a were higher than those in the observation group(P<0.05).Patients with vascular invasion of HCMC caused neuronal apoptosis,serum miR-27a expression rate was higher than TNM gradeⅠ-Ⅱ,tumor number=1,tumors≤5 cm and no vascular invasion of HCMC infected neuron apoptosis were all independent risk factors(P<0.05).After 48 hours and 72 hours of transfection,the proliferation of Caspase-3 in neuron apoptosis cells induced by HCMC infection was higher than that in the non-transfected group(P<0.05).Conclusion MicroRNA x controls HCMV virus replication through targeted inhibition,and thus plays a better antiviral role in neuronal apoptosis induced by HCMV infection,and can be used as a target point for the treatment of HCMV infection.
