摘要
目的建立HPLC法同时测定麦芽总碱提取物中3种生物碱和1种黄酮类成分含量的方法。方法色谱柱:phenomenex■C18色谱柱(250 mm×4.6 mm,5μm);流动相:甲醇-0.1 mol·L^-1磷酸二氢钾溶液,梯度洗脱;流速:1.0 mL·min^-1;柱温:30℃;进样量:20μL;检测波长:276 nm。结果辛弗林、大麦芽碱、芦竹碱、麦黄酮质量浓度分别在5~250μg·mL^-1(r=0.9997),5~250μg·mL^-1(r=0.999 9),5~250μg·mL^-1(r=0.9996)范围内线性关系良好,精密度、稳定性、重复性及加样回收率的RSD均小于3%。结论该方法操作简便,稳定性、重复性好,可用于麦芽总碱提取物中辛弗林、大麦芽碱、芦竹碱、麦黄酮的含量测定。
Objective To establish a method for simultaneous determination of three alkaloids and tricin in total malt extract.Methods HPLC assay was carried out using an phenomenex■ C18 column(250 mm×4.6 mm,5 μm)with column temperature at 30℃.The mobile phase was methanol-0.1 mol·L^-1 potassium dihydrogen phosphate solution in gradient elution at a flow rate of 1.0 mL·min^-1.And the detection wavelength was 276 nm.Results The mass concentrations of synephrine,hordenine,gramine and tricin were 5-250(r=0.999 7),5-250 μg·mL^-1(r=0.999 7),5-250 μg·mL^-1(r=0.999 9) and 5-250 μg·mL^-1(r=0.999 6),respectively.The linear relationships were good,and the RSDs of precision,stability,repeatability and recovery rates of sample addition were less than3%.Conclusion The method is simple,stable and reproducible,and can be used to determine the contents of synephrine,hordenine,gramine and tricin in totol malt extract.
