摘要
采用UHPLC法建立中药荷丹片中莲碱的含量测定方法。色谱柱为Poroshell 120 EC-C18(3.0 mm×150 mm,2.7μm),流动相为水-乙腈-三乙胺-冰乙酸(68.1∶30∶1.6∶0.3),以0.6 mL·min^(-1)流速等度洗脱,波长272 nm。结果表明:方法的专属性良好,莲碱在0.004~0.04µg范围内的绝对进样量与峰面积间具有良好的线性关系,稳定性、重复性、精密度、加样回收率的RSD均在3%以内。所建立的UHPLC法快速、准确、稳定、可靠,可作为荷丹片中莲碱的含量测定方法。
The method to determine the roemerine content in Hedan tablet was established by UHPLC.Poroshell 120 EC-C18(3.0 mm×150 mm,2.7μm)was used as the stationary phase,and the mobile phase was water-acetonitrile-triethylamine-glacial acetic acid(68.1∶30∶1.6∶0.3),equal elution was carried out with the flow rate of 0.6 mL·min^(-1),and the detection wavelength was 272 nm.The results show that,the specificity of the method was good,the absolute sample mass of roemerine from 0.004~0.04µg had a good linear relationship with the peak area.The RSD of stability,repeatability,precision and recovery were within 3%.The established UHPLC method is fast,accurate,stable and reliable,and can be used as a method for measuring roemerine content in Hedan tablet.
