摘要
目的:探讨民族药食资源栘[木衣]乙醇提取物的降糖、降脂及抗氧化活性,为其进一步资源开发提供科学依据。方法:乙醇回流法制备栘[木衣]皮的乙醇提取物,通过建立不同体外抗氧化模型1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除法、羟基自由基清除法和β-胡萝卜素-亚油酸法,以及建立体内糖脂紊乱复合小鼠模型,分别观察药物的降糖、降脂及抗氧化活性。结果:栘[木衣]皮乙醇提取物对DPPH自由基、羟基自由基以及β-胡萝卜素-亚油酸氧化体系具有明显的清除或抑制作用,且呈一定的浓度正相关;其中栘[木衣]乙醇提取物对DPPH清除作用的IC50值为147.5μg/mL,对羟基自由基清除作用的IC50值为61.11 mg/mL,对β-胡萝卜素-亚油酸法的抗氧化能力IC50值为6.308 mg/mL。同时,栘[木衣]乙醇提取物能明显或部分降低糖脂紊乱复合小鼠模型中血清FBG、TC和TG的含量,降低小鼠血清LPO和MDA的含量,升高GSH-Px及SOD活性/含量,增加GSH-Px/LPO比值,显示其具有较好的降糖、降脂活性,以及减少体内自由基损伤,降低脂质过氧化物形成的作用,增强机体抵抗氧化损伤活性。结论:栘[木衣]乙醇提取物具有一定的降糖、降脂及抗氧化活性。
Objective:To explore the hypoglycemic,lipid-reducing and antioxidant activities of the Docynia delavayi’s ethanol extract in the national medicinal resources,and provide scientific basis for further resource development.Methods:The candidate was prepared by ethanol reflux method and its antioxidant activity was observed in vitro.1,1-phenyl-2-picrylhydrazyl radical(DPPH)free radical scavenging method,hydroxyl radical scavenging method andβ-carotene-linoleic acid method were used to establish a compound glycolipid disorder mice model..Results:Docynia delavayi’s ethanol extract showed significant scavenging or inhibiting effect on hydroxyl radical,DPPH radical andβ-carotene-linoleic acid,and the effect was positively correlated with certain concentrations.The IC50 value for DPPH scavenging was 147.5 ug/mL,the IC50 value of the drug for scavenging hydroxyl radicals was 61.11 mg/mL,and the IC50 value forβ-carotene-linoleic acid method was 6.308 mg/mL.At the same time,Docynia delavayi’s ethanol extract significantly or partially reduced the serum contents of Glu,TC and TG in glycolipid disorder mice model.It also reduced the serum content of MDA,increased the activities/contents of LPO,GSH-Px and SOD,and increased the ratio of GSH-Px/LPO.It suggested that the drug had better hypoglycemic and lipid-reducing activities,in addition,the drug reduced free radical damage and the formation of lipid peroxides,and enhanced the body’s resistance to oxidative damage.Conclusion:Docynia delavayi has certain hypoglycemic,lipid-reducing and antioxidant activities.
