摘要
为研究虎杖提取物对小鼠体内抗氧化力的作用以及对果蝇的延缓衰老作用,分别以高(0.4mL)、中(0.2mL)、低(0.1mL)3个剂量虎杖提取物(质量浓度为1g/L)灌胃昆明种小鼠4周,取眼球静脉血,测定血清的总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量,分别以4个不同浓度虎杖提取物(质量分数0.1%、0.2%、0.4%、0.8%)的基本培养基连续喂养果蝇,每24h记录1次果蝇死亡数,记录果蝇存活天数.结果表明高剂量虎杖提取物显著提高了小鼠血清中的T-AOC活力,血清MDA含量也显著降低,但血清SOD活力提高不明显.质量分数0.2%虎杖提取物极显著延长雌雄果蝇寿命,对雌果蝇平均寿命延长率为31.68%,对雄果蝇平均寿命延长率为43.25%,对雌果蝇最高寿命延长率为39.05%,对雄果蝇最高寿命延长率为41.62%;添加质量分数0.4%和0.8%虎杖提取物的雌雄果蝇寿命显著延长.虎杖提取物作用于小鼠和果蝇体内,有一定的抗氧化和延缓衰老的作用.
To study the effect of the extract from Polygonum cuspidatum on antioxidant activity in mice and the anti-aging effect on Drosophila melanogaster in vivo,Three groups of Kunming mice were intragastrically administered with high(0.4mL),medium(0.2mL)and low(0.1mL)of the extract(1g/L concentration)from P.cuspidatum for 4weeks.The ocular venous blood was taken to determine the total antioxidant capacity of the serum(T-AOC),superoxide dismutase(SOD)activity and malondialdehyde(MDA)content.The D.melanogaster were continuously fed with four different concentrations of the extract from P.cuspidatum(0.1%,0.2%,0.4%,0.8%),and the number of dead D.melanogaster was recorded every 24hours,and the number of days of D.melanogaster survival was recorded.The results showed that the high dose of the extract from P.cuspidatum significantly increased the T-AOC activity in the serum of mice,and the serum MDA content also decreased very significantly,but the serum SOD activity did not increase significantly.0.2%of the extract fromP.cuspidatumsignificantly prolonged the lifespan of male and female D.melanogaster,the average life expectancy prolonging rate of female D.melanogaster was 31.68%,the average life expectancy prolonging rate of male D.melanogaster was 43.25%,and the maximum life expectancy prolonging rate of female D.melanogaster was 39.05%.The maximum life expectancy prolonging rate of male D.melanogaster was 41.62%,and the lifespan of male and female D.melanogaster with 0.4%and 0.8%of the extract from P cuspidatum was significantly prolonged.The extract from P.cuspidatum has certain anti-oxidation.anti-aging effects on mice and D.melanogaster.
