摘要
目的:基于麻黄具有"利水消肿"功效,评价麻黄对肾性水肿病症结合模型的影响,初步探讨中药性味理论。方法:采用注射用盐酸多柔比星(阿霉素)建立大鼠肾水肿模型、阿霉素以及甲状腺片建立大鼠肾阴虚水肿模型。造模成功后,模型鼠连续灌胃麻黄水煎液1. 17 g/kg、2. 34 g/kg、4. 68 g/kg 4 w。观察大鼠一般行为学、24 h尿量、24 h尿蛋白、尿素(Urea)、血浆中环磷酸腺苷(c AMP)、环磷酸鸟苷(c GMP)、血清中睾酮(T)、雌二醇(E2)、三碘甲状腺原氨酸(T3)、甲状腺素(T4)、总三酰甘油(TG)、总胆固醇(TC)、白蛋白(Alb)、总蛋白(STP)的含量来考察麻黄对肾性水肿病症结合模型的影响。结果:造模期间,大鼠出现脱毛现象且毛发光泽度较差,饮水量增多且尿量减少,尿蛋白含量增高、肾脏系数增大、血清STP、Alb含量降低,Urea、TG、TC含量升高。肾阴虚水肿组大鼠还出现大便干结,小便赤黄,狂躁易怒,E2、T3、T4含量、c AMP/c GMP值升高,T含量降低。持续给药4w后,麻黄水煎液2. 34 g/kg、4. 68 g/kg可以显著增加肾水肿模型大鼠24 h尿量、降低24 h尿蛋白以及Urea含量、增加Alb以及STP的含量、降低TG的含量和肾脏系数。麻黄水煎液各剂量显著增加肾阴虚水肿模型大鼠24 h尿量、降低24 h尿蛋白、降低血清TC、TG的含量,使T、T4含量降至更低,反而会加重肾水肿模型大鼠的狂躁易怒现象。结论:麻黄具有明显的利尿作用,对肾水肿模型大鼠有一定的治疗效果;对肾阴虚水肿模型大鼠无治疗效果,甚至会使肾阴虚的病症加重,推测其可能与麻黄的味苦辛,性温的性味功效相关。
Objects: Based on the effect of Ephedrae Herba with Lishuixiaozhong,to evaluate the effect of Ephedrae Herba on the combined model of renal edema,and do preliminary study on theory of traditional Chinese medicine. Methods: Renal edema rat model was established by injection of doxorubicin hydrochloride (doxorubicin). Shenyinxu edema rat model was established by injection of doxorubicin and thyroid tablets. After modeling,the model rats were administered with 1. 17 g/kg,2. 34 g/kg and 4. 68 g/kg Ephedrae Herba decoction for 4 weeks.General behavior of rats,24 h urine volume,24 h urine protein,urea (Urea),cyclic adenosine monophosphate (cA MP),levels of cyclic guanosine monophosphate (cG MP),testosterone (T),estradiol (E2),Triiodothyronine (T3),thyroxine (T4),serum total triglyceride (TG),serum total cholesterol (TC),serum albumin (Alb),serum total protein (STP) were observed to study the effect of Ephedrae Herba on the combined model of renal edema diseases. Results: During the modeling period,the rats developed hair loss and the gloss was poor,the water consumption was increased and the urine volume was decreased,the contents of urine protein,Urea,TG and TC were increased,the contents of STP and Alb were decreased,and the kidney coefficient was increased. In Shenyinxu edema group,dry stools,red urine and irritability were observed,the contents of E2,T3 and T4 were increased,cA MP/cG MP was increased,while T content was decreased. After4 weeks of administration,Ephedrae Herba decoction (2. 34 g/kg and 4. 68 g/kg) significantly increased the 24 h urine volume,reduced 24 h urine protein and Urea content,increased the contents of Alb and STP,reduced TG content,and reduce the kidney coefficient. In Shenyinxu edema model,all doses of Ephedrae Herba decoction significantly increased the 24 urine volume,reduced 24 h urine protein,reduced the contents fo TC and TG,more significantly reduced the contents of T and T4. It aggravated the irritability on renal edema rats. Conclusion:Ephedrae Herba has an obvious diuretic effect. It has certain therapeutic effect on renal edema rats. Ephedrae Herba has no effect on edema rats with Shenyinxu,or even aggravated the Shenyinxu symptom,which may be related to the " Weikuxin,xingwen" of Ephedrae Herba in property.
