北五味子水提物对不可预知慢性应激抑郁小鼠行为学及海马中相关神经递质的影响 被引量:9

The Influence of Water Extract of Schisandra Chinensis on Behavioral Science and Related Neurotransmitters in Hippocampus of Mice with Unpredictable Chronic Mild Stress Depression

摘要 目的:观察北五味子水提物对不可预知慢性应激(unpredictable chronic mild stress,UCMS)模型小鼠行为学、海马中相关神经递质的影响。方法:将72只小鼠按照随机数字表法分为对照组、模型组、文拉法辛组、北五味子水提物低剂量组、北五味子水提物中剂量组和北五味子水提物高剂量组,每组12只。给予UCMS加单笼单养刺激建立慢性应激抑郁小鼠模型。造模5周后进行强迫游泳测试、悬尾测试和旷场测试。行为学测试结束后,LC-MS/MS法测定小鼠海马中神经递质的含量;蛋白免疫印迹法分析小鼠海马中SERT、5HT1A蛋白表达水平。结果:模型组小鼠强迫游泳和悬尾不动时间显著增加,旷场总活动路程、中央活动路程和中央活动时间明显减少,海马中神经递质含量显著降低,海马中SERT、5HT1A蛋白表达明显下调。北五味子水提物高剂量组、中剂量组以及文拉法辛组小鼠强迫游泳和悬尾不动时间显著升高,旷场总活动路程、中央活动路程和中央活动时间明显升高,海马中神经递质含量明显上升,海马中SERT、5HT1A蛋白表达显著上调。结论:北五味子水提物对UCMS抑郁模型小鼠具有抗抑郁疗效,提高抑郁小鼠海马中神经递质含量以及相关蛋白的表达可能是其抗抑郁的作用机制之一。 Objective:To observe the influence of water extract of Schisandra chinensis on behavioral science and related neurotransmitters in hippocampus of mice with unpredictable chronic mild stress(UCMS)depression,and expression of serotonin-related protein in hippocampus.Methods:According to the random number table method,all the rats were divided into the control group,the model group,the venlafaxine group,the low-dose group of Schisandra chinensis water extract,the medium-dose group of Schisandra chinensis water extract and the high-dose group of Schisandra chinensis water extract,with 12 rats in each group.The model of chronic mild stress depression in mice was established by UCMS plus single cage monotrophic stimulation.Forced swimming test,tail suspension test and open field test were conducted after 5 weeks of modeling.After the behavioral test,the content of neurotransmitters in the hippocampus of mice was determined by LC-MS/MS,and the expression levels of SERT and 5HT1A in the hippocampus of mice were analyzed by Western blotting.Results:In the model group,forced swimming and tail suspension immobility time increased significantly,total open field activity distance,central activity distance and central activity time decreased significantly,neurotransmitter content in hippocampus decreased significantly,and expression of SERT and 5HT1A protein in hippocampus decreased significantly.The time of forced swimming and tail suspension was significantly increased in high-dose group,middle-dose group and venlafaxine groups of Schisandra chinensis water extract.The total distance of open field,central distance and central time of activity were significantly increased.The content of neurotransmitters in hippocampus was significantly increased.The expression of SERT and 5HT1A protein in hippocampus was significantly increased.Conclusion:The water extract of Schisandra chinensis has antidepressant effect on UCMS depression model mice,can increase the content of neurotransmitters and the expression of related proteins in hippocampus,which may be one of its antidepressant mechanisms.
