摘要
防御相关基因与植物抗病性密切相关,本研究以砂糖橘为实验材料,利用实时荧光定量PCR技术分析了砂糖橘CrCu/ZnSOD、CrPOD和CrPPO等防御相关基因在接种黄龙病菌(Candidatus Liberibacter asiaticus)后不同时间的动态表达差异。结果表明嫁接病芽10周后,砂糖橘叶片开始表现出黄龙病的各种典型症状。砂糖橘叶片中CrCu/ZnSOD、CrPOD和CrPPO均被显著诱导表达,表达量均呈现先升高后下降的趋势,其中CrCu/ZnSOD在嫁接病芽后20周表达水平最高,比对照组上调5.39倍,而CrPOD和CrPPO则分别在第30周和37周达到峰值,与对照相比分别上调4.7倍和1.81倍;证实砂糖橘的防御相关基因确实受到黄龙病菌侵染的诱导,从而提高其对黄龙病菌的抗性。研究结果将对柑橘抵御黄龙病菌的机制及防控探究提供帮助。
Defense-related genes are closely related to plant disease resistance.In this study,the expression of defense-related genes(CrCu/ZnSOD,CrPOD and CrPPO)were analyzed in leaves of Citrus reticulate infected by Candidatus Liberibacter asiaticus(Ca Las)at different time after inoculation by real-time quantitative PCR.Results showed that the leaves of C.reticulate appeared various Candidatus Liberibacter asiaticus typical symptoms about10 weeks after inoculation.The expression of CrCu/ZnSOD,CrPOD and CrPPO in leaves of C.reticulate were all significantly induced,and the expression levels increased firstly and then decreased.The expression levels of CrCu/ZnSOD,CrPOD and CrPPO reached the maximum at 20,30 and 37 weeks after inoculation,and were up-regulated 5.39,4.7 and 1.81 folds in comparison with the control,respectively.The results verified that the defense-related genes of C.reticulate were actually induced by the infection of Ca Las,so as to enhance its resistance to Ca Las infection.The results of this study could provide help for.the research on the mechanism and control of C.reticulate to Candidatus Liberibacter asiaticus.
