气相色谱-燃烧-同位素比值质谱法测定枸杞氨基酸δ15NAir值 被引量:7

Determination of δ15NAir of amino acids in wolfberries by gas chromatography-combustion-isotope ratio mass spectrometry

摘要 建立了以氯甲酸乙酯为衍生化试剂测定氨基酸氮稳定同位素比值的气相色谱-燃烧-同位素比值质谱法。通过对12种氨基酸进行衍生,采用TG-35MS中极性色谱柱,不分流模式进样,对应的氨基酸衍生产物可完全分离。该方法测定δ15NAir值的精密度为0. 13‰~0. 52‰,满足同位素质谱分析精度要求。与元素分析-稳定同位素质谱测定得到的δ15NAir值结果一致性较好(r2=0. 990),显示没有产生明显的同位素分馏。方法可用于枸杞中氨基酸的δ15NAir值测定并有望推广至其他领域。 A method was established to measure the nitrogen isotope of amino acids by gas chromatography-combustion-isotope ratio mass spectrometry(GC-C-IRMS).Twelve amino acids were derived with ethyl chloroformate.Amino acid derivatives were effectively separated by TG-35 MS polar column with splitless mode injection.Then,stable nitrogen isotopes were determined.The precision of GC-C-IRMS was 0.13‰-0.52‰for theδ15 NA irvalues,which fully meeted the analytical accuracy requirements of the isotopes.Besides,the results were highly correlated(R2=0.990)with the element analysis-isotope ratio mass spectrometry(EA-IRMS),indicating that no significant isotope fractionation was produced.This method can be used for theδ15 NA irdetermination of amino acids in wolfberries.
