摘要
尿苷用途广泛,发酵法是尿苷研究和生产的主流。以枯草芽孢杆菌F126为实验菌株,首先通过PB实验确定了培养基中影响尿苷产量的最显著因素是酵母浸粉。然后采用单因素实验的方法对不同厂家和型号的酵母浸粉进行了筛选,发现Springer公司所产的2506型号的酵母浸粉最有利于尿苷发酵,并确定其最佳的使用量为20 g/L。最后比较了酵母浸粉使用方法对尿苷发酵结果的影响,结果表明采用随糖流加酵母浸粉的方式更有利于尿苷的发酵生产,在最优条件下,枯草芽孢杆菌F126在5 L发酵罐上发酵40 h后可积累尿苷26.2 g/L,产量较出发条件提高了101.5%。研究结果可以为发酵法生产尿苷提供借鉴和指导。
Uridine has wide applications and fermentation is the mainstream for uridine production.Bacillus subtilis F126was chosen as the experimental strain.The most significant factor affecting uridine titer in culture medium was determined to be yeast extract by PB experiments.Single factor experiment was used to screen the best yeast extract for uridine production from different manufacturers and types.The results showed that 2506,one type of yeast extract produced by Springer,was most favorable for uridine fermentation,and the optimal dosage was 20g/L.Finally,the effects of different usages of yeast extract on fermentation were compared and it is found that feeding of yeast extract with glucose was more beneficial to uridine fermentation.Under optimal conditions,26.2g/L of uridine was accumulated after fermentation in 5Lfementator for 40hby B.subtilis F126,which was increased by 101.5%compared with that under initial conditions.The results are helpful for uridine fermentation.
