摘要
为了探索花魔芋离体培养再生微球茎体系,为魔芋产业的优良种源生产提供技术支持,本文以花魔芋愈伤组织为试材,MS培养基为基本培养基,研究不同激素组配对花魔芋愈伤组织诱导微球茎的影响。结果表明,不同激素组配和浓度水平对花魔芋愈伤组织诱导微球茎具有显著影响,当以MS为基本培养基,且转接愈伤组织培养基6-BA含量为1.0 mg/L、2,4-D含量为0.2 mg/L的配比时出球率最高(18.889%),并显著高于其他6-BA和2,4-D的配比组合;当以MS为基本培养基,且转接愈伤组织培养基中6-BA含量为1.0 mg/L、NAA含量为1.7 mg/L的配比时出球率最高(17.778%),并显著高于其他6-BA和NAA配比组合。上述结果表明,这2种不同激素的配比组合可作为花魔芋愈伤组织诱导微球茎的参选配方。
In order to explore the regeneration microcorm system,in this study,the callus was used as the test material,and MS medium was the basic medium to study the effects of different hormone combinations on the callus induced microcorms,so as to provide technical support for the good seed production of konjac industry.The results showed that the effect of different hormone combinations and concentration levels on callus induced microcorm was significant.When using MS as the basic medium,and the content of6-BA and2,4-D in the medium were1.0mg/L and0.2mg/L respectively,the ball rate was the highest(18.889%),which was significantly higher than that of other combinations of6-BA and2,4-D.While using MS as the basic medium,and the content of6-BA and NAA in the medium were1.0mg/L and1.7mg/L respectively,the ball rate was the highest(17.778%),which was significantly higher than that of other combinations of6-BA and NAA.The above results indicated that the combination of these two different hormones could be used as the recipe for the induction of microcorm by callus.
