摘要
目的分析RECQL4基因在肺癌中表达,初步功能及其与患者临床参数和预后间的关系。方法采用RT-PCR,RT-qPCR和免疫组化方法,从mRNA和蛋白水平检测RECQL4基因在正常肺、肺癌和癌旁组织中的表达。采用组织芯片免疫染色方法定量分析97例肺癌患者RECQL4蛋白的表达。RECQL4表达组间比较采用t检验,表达与临床参数:性别、年龄、临床分期、淋巴结转移、肿瘤大小和生存预后间关系采用卡方检验和Kaplan-meier分析,并通过更大样本数据库对结果进行验证。采用MTS实验分析REC-QL4基因的初步功能。结果在肺癌中RECQL4基因mRNA和蛋白水平均呈现高表达状态(P <0.01);RECQL4表达与肺癌患者临床分期(P=0.01)和肿瘤大小(P=0.025)显著相关;RECQL4低表达患者预后显著优于高表达患者(P <0.05);而且,RECQL4表达在肺腺癌患者总生存期(P <0.001)中是一个不利因素,而在肺鳞癌中RECQL4表达对患者总生存期没有影响(P=0.24)。初步功能数据发现,RECQL4基因明显促进肿瘤细胞生长和增殖,是一个潜在癌基因。结论 RECQL4是一个明显的候选癌基因和肺腺癌不良预后潜在标志物。
Objective To analyze the relationship between RECQL4 expression and clinical parameters or prognosis,and the functional roles of RECQL4 in lung cancer.Methods RT-PCR,RT-qPCR and immunohisto-chemistry were applied to detect the mRNA and protein expression of RECQL4 in normal lung,lung cancer and ad-jacent tumor tissues.The associations between RECQL4 expression and the clinical parameters such as gender,age,clinical stage,lymph node metastasis,tumor size and survival prognosis of 97 patients were analyzed using the TMA method and SPSS13.0,which was further verified by a public databases with a larger sample number.RECQL4 expressions among groups were compared using t test,and the relationship between its expression and clinicopathological characteristics or survival prognosis was analyzed using chi-square test or Kaplan-meier survival analysis.MTS experiment was used to determine the functional roles of RECQL4.Results RECQL4 was up-regu-lated in lung cancer patients both at mRNA and protein levels.A significant correlation was observed between REC-QL4 expression and clinical stages or tumor size.Finally,the patients with high RECQL4 expression showed a poor survival compared with that with low expression.Moreover,RECQL4 expression was an unfavorable prognostic marker for patients in lung adenocarcinoma,whereas RECQL4 expression had no effect on the survival of patients in lung squamous carcinoma.The functional study showed that RECQL4 significantly promoted cancer cell growth and proliferation in lung cancer.Conclusion RECQL4 is a potential oncogene and an unfavorable prognostic marker for lung adenocarcinoma patients.
