牙髓干细胞来源的外泌体对于巨噬细胞极化作用影响的实验研究 被引量:5

摘要 目的:研究牙髓干细胞(DPSCs)的外泌体(EXO)对巨噬细胞极化的影响。方法:培养SD大鼠牙髓干细胞(r DPSCs),检测相应的间充质干细胞表面标志物其多向分化潜能。提取r DPSCs的EXO,构建皮肤缺损模型,检测EXO处理的皮肤创伤组织中巨噬细胞向不同亚型极化相关因子的表达。结果:r DPSCs表达间充质干细胞标志物,具有多向分化潜能,经EXO处理的大鼠皮肤创伤组织中,TNF-α、TNF-1β、IL-6表达均下降(P <0. 05),IL-4、IL-10、IL-13表达均上升(P <0. 01)。结论:r DPSCs分泌的EXO能够促进巨噬细胞向M2型极化,可能促进皮肤组织的修复再生。 Objective:To study the influence of exosome(EXO)secreted by the rat dental pulp stem cells(rDPSCs)on macrophage polarization.Methods:rDPSCs were cultured from SD rats,the immunophenotype and multi-directional differentiation potential were detected.Skin wound model was created and treated with EXO.The macrophage polarization were observed by the detection of polarization related gene expression by qPCR.Results:rDPSCs positively expressed mesenchymal stem cell markers and showed multiple differentiation potential.The EXO treated macrophages decreased the expression of TNF-α,TNF-1βand IL-6(P<0.05),and increased the expression of IL-4,IL-10 and IL-13(P<0.01).Conclusion:EXO secreted by rDPSCs can promote the polarization of macrophages towards anti-inflammatory phenotype(M2)and may promote the repair and regeneration of wounded skin tissue.
