一株产碱性CMC酶链霉菌的筛选及酶学性质研究 被引量:3

摘要 经CMC平板分离筛选,从青藏高原牦牛粪样品中获得一株产碱性CMC酶能力较强的菌株Tibet-YD4524-3,16S rRNA基因序列初步鉴定其为链霉菌类(Streptomyces sp.)菌种.生物学特性的研究表明,Tibet-YD4524-3产酶的最佳碳源为可溶性淀粉和羧甲基纤维素钠,最佳氮源为酵母膏,最适产酶温度为30℃,最适产酶p H值为8.0.酶学性质初步研究显示,Tibet-YD4524-3产生的CMC酶最适反应温度40℃,最适反应p H值为8.5,在碱性条件下CMC酶具有较高的酶活性和较好的稳定性.金属离子K^+、Mn^(2+)和Fe^(2+)对Tibet-YD4524-3所产CMC酶的反应具有促进作用,金属离子Cu^(2+)、Pb^(2+)和Hg^(2+)具有抑制CMC酶反应的作用. By using the CMC plate screening method,an alkaline cellulase producing strain,designated as Tibet YD4524 3,was isolated from the yak dung in the Qinghai Tibet Plateau.The 16S rDNA sequence analysis classified Tibet YD4524 3 as Streptomyces sp.This strain was characterized to utilize soluble starch and CMC Na as the optimal carbon source and yeast extract as the optimal nitrogen source for cellulase producing.The optimal temperature and pH of strain Tibet YD4524 3 for cellulase producing were 30℃and pH 8.0.The cellulase produced by the strain was shown to be a kind of alkaline cellulase with the optimal catalytic activity at 40℃and pH 8.5.K+,Mn 2+,and Fe 2+were the activators while Cu 2+,Pb 2+,and Hg 2+were the inhibitors of the enzyme.
