败酱及其近缘种的分子鉴定及亲缘关系研究 被引量:1

Molecular authentication and phylogenetic relationship of Patrinia scabiosifolia and its relative species

摘要 为了构建败酱Patrinia scabiosifolia与近缘种之间系统发育关系,准确鉴别败酱及其近缘种,对其核基因ITS片段和叶绿体基因psbA-trnH序列扩增和测序,计算物种种内、种间Kimura 2-parameter,(K2P)遗传距离,并采用最近距离、相似性搜索和构建Neighbor-joining,(NJ)系统聚类树3种方法进行鉴定分析。结果表明,ITS和psbAtrnH分子系统树支持败酱不同居群样本聚为一单系分支,支持率分别为65%和68%;其中,败酱与攀倒甑(P. villosa)构成一单系分支,ITS和psbA-trnH数据的支持率分别为51%和99%。败酱及其近缘种种间ITS和psbA-trnH序列的遗传距离为0. 008 31~0. 072 92和0. 005 86~0. 039 18,均远大于败酱种内遗传距离0~0. 003 31和0。由此可知,中药材败酱与攀倒甑亲缘关系最近,ITS和psbA-trnH序列可以准确鉴别败酱及其近缘种。 To construct the phylogenetic relationships of Patrinia scabiosifolia and its relative species,and accurately identify P.scabiosifolia and its relative species nuclear DNA internal transcibed spacer(ITS)and chloroplast gene psb A-trn H sequences were amplified and sequenced.To calculate the Kimura 2-Parameter(K2P)distances.Molecular authentication analyses were performed using Nearest Distance,BLAST1 and Neighbor-joining(NJ)methods.The results showed that the NJ trees(ITS and psb A-trn H data)indicated that different populations of P.scabiosifolia formed a monophyletic clade[Bootstrap(BS)=65%and 68%].P.scabiosifolia and P.villosa formed one monophyletic clade(ITS and psb A-trn H data:BS=51%;99%).The inter-specific genetic distances(ITS and psb A-trn H data)between P.scabiosifolia and its relative species were(0.008 31~0.072 92)and(0.005 86~0.039 18),which were far higher than the intra-specific genetic distances of P.scabiosifolia(0~0.003 31)and 0.It can be seen that the closest relative of P.scabiosifolia is P.villosa.ITS and psb A-trn H are two efficient barcodes for the authentication of P.scabiosifolia and its relative species.
