摘要
为探讨外源钙对干旱胁迫下樟子松苗木生理指标的影响,本研究对干旱胁迫处理的樟子松苗施加外源钙,采用室内控制实验,分析了苗木含水量、叶绿素各组分含量、O_2^-产生速率和H_2O_2含量、抗氧化酶系统(CAT、POD和SOD)、MDA和游离脯氨酸含量变化。结果表明:干旱胁迫下,樟子松苗木含水率呈现下降趋势,施加外源钙可以有效缓解苗木含水率的下降,保护苗木抵御干旱;樟子松苗木针叶O2-产生速率和H_2O_2含量在干旱胁迫下显著升高,施加外源钙可以诱导苗木抗氧化酶系统(CAT、POD和SOD)酶活性升高,祛除因干旱胁迫造成积累的活性氧,降低活性氧对樟子松苗木的破坏程度。正常水分条件下,外源钙可以提高樟子松苗木根和茎的CAT、POD和SOD活性,但无法扭转干旱胁迫给樟子松苗木带来的伤害;与此同时,酶的活性变化与干旱胁迫的诱导密切相关。正常水分条件下,无论是否施加外源钙,均不能使樟子松苗木根和茎内的POD和SOD活性有大幅升高,具体原因有待进一步探讨。MDA的研究结果显示,外源钙可以有效降低干旱胁迫下樟子松苗木MDA含量,缓解干旱胁迫对樟子松苗木细胞造成的损害;但这种缓解依然是不能逆转。施加外源钙可以有效降低樟子松苗木游离脯氨酸含量,降低干旱胁迫对樟子松苗木细胞膜脂的过氧化破坏程度;干旱胁迫可以降低樟子松苗木光合色素各组分的含量,进而抑制苗木光合作用,而施加外源钙可以有效缓解干旱胁迫对樟子松苗木光合作用的抑制作用,保证苗木的正常生长。
In order to reveal the effects of exogenous calcium on the active oxygen metabolism system of Pinus sylvestris var.mongolica seedling needles under drought stress,and then to provide scientific basis for the research on stress-tolerance related physiology and seedling of P.sylvestris var.mongolica.The water content in P.sylvestris var.mongolica seedling under drought stress were measured with the drying weighing method.The changes of chlorophyll content in P.sylvestris var.mongolica seedlings were studied.The changes of H2O2 content,the antioxidant enzyme system(CAT,POD and SOD),and the MDA and free proline content were also studied with the plant physiology and biochemistry methods.The water content of seedling was decreasing with the degree of drought stress,and the application of external Ca could effectively alleviate the decline of water content of seedling and protect the seedling.There was insignificant change in moisture content in normal water supply treatment.O2-production rate and H2O2 content of seedling were significantly increased under drought stress,introduction of exogenous Ca on seedlings could induce the increment of enzyme(like CAT,POD and SOD)activities in antioxidant enzyme system,and eliminate accumulation of reactive oxygen caused by drought stress.The application of external calcium could accelerate the increment of antioxidant enzyme in seedlings.Through the analysis of the content of MDA and free proline in seedlings,the results indicated that the application of exogenous Ca could effectively reduce the content of MDA and free proline in seedlings,and decrease the degree of peroxidation of lipid of seedlings.The drought stress can reduce the photosynthetic pigment content of P.sylvestris var.mongolica seedlings,and inhibit the photosynthesis of seedlings,then affect the normal growth of seedlings.The application of exogenous Ca can effectively alleviate the inhibition effect of drought on photosynthesis of seedlings and ensure the normal growth of seedlings.
