摘要
以长白山野生红姑娘茎叶为原料,总黄酮得率为响应值,利用单因素和响应面法优化红姑娘茎叶总黄酮提取工艺,并评价其体外抗氧化活性。结果显示,红姑娘茎叶总黄酮提取的最佳条件为:乙醇浓度72%,料液比1∶28(g/mL),温度67℃,时间3.8 h,总黄酮得率最高为6.35 mg/g。纯化后的野生红姑娘茎叶总黄酮对超氧阴离子、DPPH·和羟基自由基的清除作用较强,同时也具备较强的还原力。野生红姑娘茎叶中的总黄酮表现出较强的体外抗氧化活性,可以作为一种天然抗氧化剂被开发利用。
The stems and leaves of Physalis alkekengi from Changbai Mountain were used as raw materials.The yield of total flavonoids was taken as the response value.Single factor and response surface method were used to optimize the extraction process of total flavonoids from stems and leaves of Physalis alkekengi and evaluate its antioxidative activity in vitro.The results showed that the optimal conditions for extraction of total flavonoids from stems and leaves of Physalis alkekengi were as follows:ethanol volume fraction 72%,solid-liquid ratio 1∶28(g/mL),temperature 67℃,time 3.8 h,and the highest yield of total flavonoids was 6.35 mg/g.After purification,the total flavonoids in stems and leaves of Physalis alkekengi had strong scavenging effects on hydroxyl radicals,superoxide anion and DPPH·,as the same time,the reducing power was also strong.The strong in vitro antioxidant activity of total flavonoids in stems and leaves of Physalis alkekengi can be exploited as a natural antioxidant.
