摘要
目的探讨铬镁联合补充对糖耐量异常(IGT)人群血糖的影响,观察循环miRNA表达谱的变化,筛查表达差异miRNA。方法选取IGT患者及与其年龄相匹配的糖耐量正常人群。分为:正常对照组、安慰剂对照组、铬镁干预组,干预3个月。测定血糖、胰岛素。二代测序(NGS)技术检测循环miRNA的表达,qPCR技术验证NGS的结果。生物信息学的方法分析差异miRNA相关的信号通路。结果干预后,铬镁干预组FBG、2h PG较安慰剂对照组下降(P<0.05),胰岛素抵抗改善。NGS测序得到了6个差异表达的循环miRNA。qPCR结果表明,miR-182、miR-96-5p和miR-27b的表达趋势与NGS相同。miR-182-5p靶基因的功能富集分析发现与之相关的信号通路是葡萄糖结合通路。与miR-96-5p靶基因相关的信号通路是磷脂酰肌醇信号通路。结论联合补充微量元素铬镁降低IGT患者的血糖,改善胰岛素抵抗。IGT患者miR-182-5p和miR-96-5p表达降低,补充铬镁后可使miR-182-5p和miR-96-5p表达升高。
Objective To investigate the effect of chromium(Cr)and magnesium(Mg)supplementation on blood glucose and circulating miRNA level in patients with impaired glucose tolerance.Methods IGT patients and their age-matched healthy population with normal glucose tolerance were divided into three groups:the healthy control group,placebo control group,Cr and Mg intervention group.Blood glucose,insulin andcirculating miRNA level were determined.Results After 3-month intervention,FBG,2h-PG in Cr-Mg intervention group were signif-icantly decreased compared with placebo control group(P<0.05),with markedly improved insulin resistance.Six differentially expressed circulating miRNAs were observed.RT-qPCR results showed that miR-182,miR-96-5p and miR-27b expression were as the same as NGS.Functional enrichment analysis of the miR-182-5p tar-get gene revealed that the glucose-binding pathway may be involved.And miR-96-5p target genes were included in phosphatidylinositol signaling pathway.Conclusion Supplementation of Cr and Mg can improve glucose tolerance of IGT patients.After supplementation with Cr and Mg,levels of miR-182-5p and miR-96-5p were increased.
