不同有机氮源对发酵法生产肌苷影响的研究 被引量:4

摘要 以枯草芽孢杆菌(Bacillus subtilis)HX1026为研究对象,研究大豆蛋白胨、酵母粉、玉米浆干粉和豆粕水解液4种有机氮源对发酵法生产肌苷产量的影响。结果表明,以豆粕水解液为有机氮源时,肌苷产量高于其他有机氮源,其中山东1号豆粕水解液摇瓶发酵肌苷产量最高,达到31.2g/L。在20L发酵罐验证实验中,肌苷产量达到51.2g/L,发酵罐中肌苷晶体的形成过程清晰可见,进一步证实山东1号是最适合B.subtilis HX1026发酵生产肌苷的有机氮源。山东1号的主要技术参数如总氮含量、氨基氮含量、氨基氮/总氮比值分别是3.22%、2.06%、0.639,可以作为肌苷发酵生产过程中选择有机氮源的标准,以确保肌苷发酵过程的稳定性。 Using Bacillus subt^ilis HX1026 as the research object,the effects of four organic nitrogen sources such as soybean peptone,yeast powder,corn steep liquor dry powder and soybean meal hydrolysate on the production of inosine by fermentation were investigated.The results showed that when soybean meal hydrolysate was used as organic nitrogen source,the inosine yield was higher than that of using other organic nitrogen sources,among which,the yield of inosine by Shandong No.1 soybean meal hydrolysate shaker fermentation was the highest,reaching 31.2 g/L.In the 20 L fermenter verification experiments,the inosine yield was up to 51.2 g/L,and the formation of inosine crystals in fermenter was clearly visible,which was further confirmed that Shandong No.1 was the most suitable organic nitrogen for the production of inosine by B.subtilis HX1026 fermentation.The main technical parameters of Shandong No.1 such as total nitrogen,amino nitrogen,the ratio of amino nitrogen and total nitrogen were 3.22%,2.06%,0.639,respectively,which could be used as the standard of organic nitrogen source in the production process of inosine fermentation to ensure the stability of the inosine fermentation process.
