摘要
以DPPH自由基清除活性为指标对青蛤酶解条件(即时间、温度、pH、酶添加量和固液比)进行正交试验设计,研究了青蛤蛋白酶解物的抗氧化活性。结果表明,酶解条件为温度40℃、pH 11.5、酶添加量3000U/g原料以及固液比为1∶2(w/w)时,酶解物的DPPH自由基的清除活性最高。对最佳水解条件下所获得的酶解物进行抗氧化活性测试,内容包括DPPH自由基清除活性、超氧阴离子清除活性、羟基自由基清除活性和还原能力。结果显示,酶解物有较低超氧阴离子清除活性,但却具有较高DPPH自由基清除活性、羟基自由基清除活性和还原能力。
The hydrolysates were assessed using methods of DPPH radical scavenging ability.The antioxidant properties of enzymatically hydrolyzed Cyclina sinensis were studied.Hydrolysis conditions(viz.,temperature,pH enzyme to substrate level and solid/liquid ratio)for preparing protein hydrolysates from the yclina sinensis meal proteins were optimized by orthogonal design.The results were that temperature of 40℃,pH 11.5,an enzyme to substrate level of 3000U/g,and ratio of solid/liquid ratio of 1∶2(w/w),were found to be the optimum conditions to obtain the best antioxidative hydrolysates.In the best hydrolysis conditions,antioxidant activities,DPPH radical scavenging activity including reducing power,superoxide anion and hydroxyl radical scavenging activity,of the hydrolysates were evaluated.The results showed that the hydrolysates of Cyclina sinensis exhibited the lower superoxide anion radical scavenging activities,but the higher reducing powers,DPPH radical scavenging activities and Hydroxyl radical scavenging activities.
