摘要
目的探讨大鼠肺缺血再灌注损伤中阿米洛利的作用机制。方法选择雄性实验大鼠72只作为研究对象,随机分成三组,每组24只,分别为IR组(缺血再灌注)、假手术组(C组)、阿米洛利组(A组),分别在缺血45 min、再灌注60 min和120 min 3个不同时点将各组实验大鼠麻醉处死后取血浆和右肺组织,并对其进行检测。结果 IR组再灌注60、120 min时,丙二醛(MDA)、凋亡指数(AI)较假手术组均有明显升高,阿米洛利组在给予药物后,MDA、AI与IR组比较均有明显降低(P<0.05)。与假手术组相比,IR组各个时点的肺组织湿/干质量比(W/D)和髓过氧化物酶(MPO)活性均显著增高(P<0.05),而阿米洛利给药后肺组织W/D和MPO活性则显著降低。结论阿米洛利对肺缺血再灌注损伤具有很强的保护作用,这可能是通过减少钙离子,从而清除体内过量的氧自由基,达成抑制中性粒细胞浸润和减少细胞凋亡的目的。
Objective To explore the mechanism of effects of Amiloride on rats with pulmonary ischemia reperfusion injury.Methods Seventy-two experimental male rats were selected and randomly divided into the IR group(the ischemia reperfusion group),the sham operation group(group C)and the Amiloride group(group A),24 rats in each group.After 45 min ischemia,60 min reperfusion and 120 min,rats in each group were anesthetized and killed,then plasma and right lung tissues were taken and detected.Results At 60 and 120 min reperfusion,malondialdehyde(MDA),apoptosis index(AI)in the IR group were significantly higher than those in the sham operation group.The Amiloride group was given drugs,MDA and AI were significantly lower,compared with the IR group(P<0.05).Compared with the sham operation group,the lung wet/dry weight ratio(W/D)and myeloperoxidase(MPO)activity in the IR group at each time point were significantly increased(P<0.05),while Amiloride after administration,the W/D in lung tissue and MPO activity decreased significantly.Conclusion Amiloride on lung ischemia reperfusion injury has a strong protective effect,this may be by reducing calcium ion,so as to remove the body of excess oxygen free radicals and inhibition of neutrophil infiltration and reduce cell apoptosis of purpose.
