摘要
目的研究甘草酸软膏对慢性湿疹小鼠模型的免疫学机制的影响。方法小鼠随机分为空白对照组、模型对照组、基质对照组、醋酸地塞米松乳膏组、甘草酸软膏大、中、小剂量组,除空白对照组外其他组分别建立2,4-硝基氯苯反复刺激的慢性湿疹小鼠模型,给药14 d后检测血清白细胞介素(IL)-2、IL-4、IL-5、IL-6、IL-18和干扰素(IFN)-γ含量及对外周血中T细胞亚群进行测定。结果甘草酸软膏大、中剂量组能降血清IL-2、IL-5、IL-6、IL-18和IFN-γ的含量,与模型对照组比较,差异有统计学意义(P<0.01或P<0.05);甘草酸软膏大、中剂量组能升高血清IL-4的含量,与模型对照组比较,差异有统计学意义(P<0.01或P<0.05)。甘草酸软膏大、中剂量组CD_4^+百分比和CD_4^+/CD_8^+明显升高,而CD_8^+百分比明显降低,与模型对照组比较,差异有统计学意义(P<0.01或P<0.05)。结论甘草酸软膏可通过调节Th1和Th2平衡,纠正免疫功能失调而达到治疗慢性湿疹的目的。
ObjectiveTo observe the immunological mechanism of glycyrrhizic acid ointment on chronic eczema mouse model.MethodsMice were randomly divided into blank control group,model control group,base material group,dexamethasone acetate cream group,glycyrrhizic acid ointment high,medium and low dose groups.Except for the blank control group,chronic eczema mouse model was established by chronic repeated2,4nitrochlorobenzene stimulation.The contents of IL4,IL18,IFNγin serum and the T lymphocyte subpopulation in the peripheral blood were all determined after14days.ResultsGlycyrrhizic acid ointment in high,medium groups could significantly reduce the contents of interleukin(IL)2,IL5,IL6,IL18,interferon(IFN)γin serum,with significant differences as compared with the model control group(P<0.01or P<0.05).The content of IL4in serum was significantly increased in the glycyrrhizic acid ointment high,medium groups as compared with the model control group(P<0.01or P<0.05).Compared with model control group,CD+4T lymphocyte percentage and CD+4/CD+8T in peripheral blood were significantly increased,and CD+8T lymphocyte percentage decreased significantly(P<0.01or P<0.05).ConclusionGlycyrrhizic acid ointment's effect of treating chronic eczema may be related to regulating the balance of Th1and Th2and correcting the immune disorders.
