摘要
目的研究靶向METCAM/MUC18的siRNA在肝癌细胞迁移、侵袭中的作用。方法培养肝癌细胞株Hep G2并转染MUC18的siRNA和阴性对照(NC)的siRNA,转染siRNA后检测细胞的迁移和侵袭数目、MMPs及上皮间质转化标志基因的m RNA表达量。结果 MUC18-siRNA组Hep G2细胞中MUC18的m RNA表达量显著低于NC-siRNA组;转染siRNA后12 h、24 h,MUC18-siRNA组Hep G2细胞的迁移数目和侵袭数目均显著低于NC-siRNA组;转染siRNA后24 h,MUC18-siRNA组Hep G2细胞MMP8、MMP9、MMP14、N-cadherin、Vimentin的m RNA表达量均显著低于NC-siRNA组,E-cadherin的m RNA表达量显著高于NC-siRNA组。结论靶向METCAM/MUC18的siRNA能够通过抑制MMPs表达和上皮间质转化来抑制肝癌细胞的迁移、侵袭过程。
Objective To study the regulation effect of siRNA targeting METCAM/MUC18on the migration and invasion of hepatocellularcarcinoma cells.Methods HepG2cells were cultured and transfected with MUC18-siRNA,negative control(NC)siRNA.After transfectionof siRNA,migration and invasion numbers of cells,mRNA expression of MMPs,and epithelial mesenchymal transition marker gene weredetermined.Results mRNA expression of MUC18in HepG2cells of MUC18-siRNA group was significantly lower than the NC-siRNAgroup;12hours and24hours after the transfection of siRNA,migration and invasion numbers of HepG2cells of MUC18-siRNA groupwere significantly lower than the NC-siRNA group;24hours after the transfection of siRNA,mRNA expression of MMP8,MMP9,MMP14,N-cadherin,Vimentin in HepG2cells of MUC18-siRNA group were significantly lower than the NC-siRNA group,mRNA expression ofE-cadherin was significantly higher than that of NC-siRNA group.Conclusion siRNA targeting METCAM/MUC18can inhibit the migrationand invasion of hepatocellular carcinoma cells by inhibiting MMPs expression and epithelial mesenchymal transition.
