A型肉毒毒素对人瘢痕疙瘩成纤维细胞凋亡影响的实验研究 被引量:6

摘要 目的:探讨A型肉毒毒素对人瘢痕疙瘩成纤维细胞凋亡的影响。方法:体外分离和培养人瘢痕疙瘩成纤维细胞,实验组在细胞培养过程中加入A型肉毒毒素(稀释成5U/0.1ml)进行干扰,空白对照组培养液中不加A型肉毒毒素。培养72h后利用流式细胞仪检测细胞凋亡情况。结果:3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐[3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide,MTT]法显示A型肉毒毒素作用后细胞增殖速度变慢;流式细胞仪检测显示实验组早期凋亡和晚期凋亡的细胞数量较空白对照组均明显增多,成纤维细胞凋亡率差异有统计学意义(P<0.05)。结论:A型肉毒毒素可以抑制瘢痕疙瘩组织中成纤维细胞的增殖活性,诱导成纤维细胞的凋亡。 Objective To investigate the effects of botulinum toxin type A(BTXA)on apoptosis of human keloid fibroblast.Methods Human keloid fibroblast was isolated and cultured in vitro,the medium containing BTXA(diluted concentration of5U/0.1ml)was used to intervene the cell growth in experience group,BTXA was not added in blank control group.At72hours after cells were cultured,cells apoptosis were measured by flow cytometry(FCM).Results Cells proliferation rate was slow with MTT method.The cell numbers of early and late apoptosis in experience group were significantly more than those of early and late apoptosis in blank control group with FCM,showing a significant difference between them(P<0.05).Conclusion BTXA can inhibit the proliferation and induce apoptosis of human keloid fibroblast.
