三氧化二砷脂质体的制备及银盐法含药量测定 被引量:1

摘要 目的:制备三氧化二砷脂质体,并建立测定三氧化二砷脂质体药物含量及包封率的方法。方法:通过醋酸铜梯度法制备三氧化二砷脂质体,利用银盐法测定砷的含量,采用葡聚糖凝胶微柱离心法分离游离药物与脂质体,并测定包封率。结果:确定银盐法检测砷的波长为510 nm,线性范围为4. 0~24. 0μg(r=0. 999 1),精密度、回收率符合要求。通过葡聚糖凝胶微柱离心可完全分离脂质体和游离药物。脂质体中砷含量为0. 296 mg·m L-1,包封率为83. 54%。结论:该方法准确度高、重复性好,适用于三氧化二砷脂质体的质量评价。 Objective: To prepare arsenic trioxide liposomes, and establish a method to determine the arsenic content and encapsulation efficiency of liposomes. Methods: The arsenic trioxide liposomes were prepared by copper acetate transmem- brane gradients. The content of arsenic was determined according to the silver diethyl-dithiocarbamate method. A sephadex micro- column centrifugation method was used for the separation of free arsenic trioxide from liposomes, and applied for the determination of encapsulation efficiency. Results: The determination wavelength was colffirmed to be 510 nm and the calibrated linear curve of elemental arsenic was in the range of 4.0-24.0 μg( r =0. 999 l ). The precision and recovery was accorded with the requirement of methodology. The free drug was completely separated from liposomes by centrifuging through sephadex microcol- umn. The content of arsenic in arsenic trioxide liposomes suspension was 0. 296 mg .mL-1 and the entrapment efficiency was 83. 54%. Conclusions: The method is accurate and reliable, which can be used to evaluate the quality of arsenic trioxide liposomes.
