摘要
目的研究意大利牛舌草提取物对大鼠离体胸主动脉的舒张作用。方法将意大利牛舌草粉碎成粗粉,70%乙醇超声提取3次,浓缩提取液。处死大鼠,分离胸主动脉,制作血管环,分别设置内皮完整组和内皮去除组,2组累积加药法记录牛舌草提取物溶液(1.0、3.0、5.0、7.0、9.0、11.0mg/mL)对去甲肾上腺素(PE)预收缩的胸主动脉环的张力的影响,测定血管舒张率。结果牛舌草对大鼠内皮完整组和内皮去除组的胸主动脉环均具有浓度依赖性的舒张作用,内皮完整组和内皮去除组的Emax分别为(93.82%±5.35%)和(91.54%±5.00%),2组舒张作用效果相比较,差异无统计学意义(P>0.05)。结论牛舌草对大鼠胸主动脉环具有剂量依赖性舒张血管的作用,其舒张血管作用与血管上的内皮细胞无依赖作用。
Objective To investigate the relaxation effect of extract of Anchusa italica Retizon isolated thoracic aorta in rats. Methods The Anchusa italica Retiz was pulverized into coarse powder, followed by ultrasonical extraction with 70% ethanol for 3 times, and condensed the extract. The rat thoracic aorta rings were prepared by means of isolated vascular ring, which were randomly divided into endothelium-denuded rings group and endothelium-intact rings group. The isometric tension of aortic rings induced by phenylephrine （PE） with or without endothelium was measured in solutions contained the extract of Anchusa italica Retiz （1.0, 3.0, 5.0, 7.0, 9.0, 11.0 mg/mL）. Results The extract of Anchusa italica Retiz produced concentration-dependent, relaxations in PE-contracted aortic rings with or without endothelium. The maximum relaxation by the extract of Anchusa italica Retiz was 93.82%±5.35% in endothelium intact rings and 91.54%±5.00% in endothelium removed rings, there was no significant difference between the two groups （P 〉0.05）. Conclusion／ Anchusa italica Retiz. can give rise to the concentration-dependent endothelium-independent relaxations in rat thoracic aortic rings.
