乙酰化鼠尾草酸的稳定性及对海马神经元的保护作用 被引量:2

Stability of acetylated carnosic acid and its protective effect on hippocampal neurons

摘要 目的:在确保化合物稳定性的前提下,评价乙酰化鼠尾草酸对小鼠海马神经元的保护作用。方法:C57小鼠胎鼠海马神经元经乙酰化鼠尾草酸5、20、80μmol/L预处理后,加入H_2O_2,抗体染色,观察细胞状态并评估神经元的活性。同时,高效液相色谱法测定168 h内四种不同保存条件下乙酰化鼠尾草酸的含量。结果:乙酰鼠尾草酸预处理组与模型组相比,小鼠海马神经元活性增强,LDH的漏出量下降,细胞内MDA产生下降,SOD活性升高,caspase-3 mRNA的表达降低;在168 h内,乙酰鼠尾草酸在四种不同溶液保存条件下,含量稳定。结论:乙酰鼠尾草酸在不同溶液中结构性质稳定,对小鼠海马神经元的损伤具有良好的保护作用。 Objective: To evaluate the effect of acetylated carnosic acid on hippocampal neurons. Methods: Neurons were treated with acetylated carnosic acid at concentrations of 5,20,80μmol/L for 30 min prior to H_2O_2(50. 0 μmol/L) treatment. After 6 h,the morphology and activity of neurons were evaluated. Meanwhile,contents of acetylated carnosic acid were detected by HPLC in 0 ~ 168 h under different storage conditions. Results: The activity of hippocampal neurons increased after the pretreatment with acetylated carnosic acid. The content of acetylated carnosic acid kept stable. Conclusion: Acetylated carnosic acid can protect injured hippocampal neurons induced by H_2O_2,and the compound keeps stability well.
