摘要
【目的】观察升清降浊胶囊对5/6肾切除大鼠肾功能和炎症损伤的影响,探讨其对肾脏的保护机制。【方法】采用5/6肾切除法复制慢性肾脏病(CKD)大鼠模型。最终大鼠被分别纳入正常对照组(n=6)、假手术组(n=6)、模型组(n=7)和低、中、高剂量治疗组(每组n=6)。低、中、高剂量治疗组分别给予剂量为400、800、1 200 mg·kg^(-1)·d^(-1)的升清降浊胶囊灌胃,正常对照组、假手术组、模型组给予等量生理盐水灌胃。干预8周后,检测各组大鼠血肌酐(SCr)、尿素氮(BUN)、24h尿蛋白(24h UP)定量,观察大鼠肾组织病理形态学变化,并采用酶联免疫吸附(ELISA)法测定大鼠血清白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)水平。【结果】与假手术组比较,模型组大鼠血清SCr、BUN、24h UP显著升高(P<0.01),肾小球硬化和肾小管间质纤维化明显,IL-1β、IL-6、TNF-α水平显著升高(P<0.01);与模型组比较,除低剂量组BUN、24h UP无明显变化外,3个剂量治疗组SCr、BUN、24h UP明显降低(P<0.01),肾小球硬化和肾小管间质纤维化有不同程度的改善,IL-1β、IL-6、TNF-α表达水平均明显下降(P<0.05或P<0.01),呈剂量依赖性。【结论】升清降浊胶囊能有效改善CKD大鼠肾功能和肾纤维化,其途径可能与下调炎症因子IL-1β、IL-6、TNF-α水平,从而减轻肾脏炎症损伤有关。
Objective To observe the effects of Shengqing Jiangzhuo Capsules（SJC）on renal function andinflammatory injury in 5/6 nephrectomied rats,and to investigate its renal protective mechanism. Methods The ratmodel of chronic kideny disease（CKD）was established by 5/6 nephrectomy. The rats were enrolled in normalcontrol group（n = 6）,sham-operation group（n = 6）,model group（n = 7）,and low-,middle-and high-dosetreatment groups（6 rats in each group）,respectively. Low-,middle-and high-dose treatment groups were givenintragastric administration of SJC at the dosage of 400,800,1 200 mg·kg（-1）·d（-1）,respectively,and the normalcontrol group,sham-operation group,and model group were given intragastric administration of the same volumeof normal saline. After 8-week treatment,serum creatinine（SCr）,blood urea nitrogen（BUN）and 24-h urineprotein（24 h UP）were detected,the pathological features of the renal tissues were observed,and the serum levelsof interleukin（IL）-1β, IL-6 and tumor necrosis factor α（TNF-α） were determined by enzyme-linkedimmunosorbent assay（ELISA）. Results Compared with the sham-operation group, the serum levels of SCr,BUN,24 h UP in the model group were increased,glomerular sclerosis and renal tubule interstitial fibrosis were obvious,and the levels of IL-1β,IL-6 and TNF-α were increased（P 〈 0.01）. Compared with the model group,SCr,BUN,24 h UP were decreased,the renal tubule interstitial fibrosis were improved,and IL-1β,IL-6 andTNF-α expressions levels were decreased in the 3 treatment groups（P 〈 0.05 or P 〈 0.01）except that BUN and 24 h UP levels in the low-dose SJC group had no obvious changes,the effect being dose-dependent. Conclusion SJC has certain effects on improving the renal function and renal fibrosis in CKD rats,and its mechanism is relatedto the down-regulation of the levels of inflammatory cytokines IL-1β,IL-6,TNF-α,and with the relief of renalinflammatory injury.
