摘要
目的探讨麝香配伍乳香对正常C57BL/6小鼠前列腺组织Claudins mRNA表达的影响。方法正常C57BL/6小鼠48只随机分为4组:麝香组、乳香组、配伍组和对照组,每组12只。按组别分别给予麝香、乳香、麝香+乳香、生理盐水灌胃,分别于24、48h时处死6只取前列腺组织。采用实时荧光定量PCR方法检测小鼠前列腺组织Claudin-1、Claudin-2、Claudin-3、Claudin-4、Claudin-5、Claudin-7、Claudin-8、Claudin-10mRNA表达。结果 24h时,与对照组比较,配伍组Claudin-2、Claudin-3、Claudin-4、Claudin-8表达下调(P<0.05,P<0.01);与麝香组和乳香组比较,配伍组Claudin-2、Claudin-3、Claudin-4、Claudin-5、Claudin-8表达下调(P<0.05,P<0.01)。48h时,与对照组比较,麝香组Claudin-2、Claudin-3、Claudin-4、Claudin-5、Claudin-8mRNA表达下调(P<0.05,P<0.01),乳香组Claudin-5 mRNA表达下调(P<0.05),配伍组Claudin-5mRNA表达下调(P<0.05);与配伍组比较,麝香组Claudin-2、Claudin-8 mRNA表达下调(P<0.05,P<0.01)。结论麝香配伍乳香可加快Claudin-2、Claudin-3、Claudin-4、Claudin-8mRNA表达下调速度,两种药物具有"相须"为用的协同性。
Objective To investigate the effects of Moschus moschiferus L.combined with Frankincense on regulation of Claudins mRNA expression in prostate of C57BL/6 mice. Met hod Totally 4 8 C57 BL/6 mice were randomly divided into 4 groups, which were the Moschus moschiferus L.（musk） group,Frankincense group,combination group and control group,12 in each group. The m ice were given with Moschus moschiferus L..,Frankincense, Moschus m oschiferus L. combined with Frankincense,and saline by gavage respectively. Six mice in each group were sacrificed after 2 4 and 4 8 hours respectively. Expressions of Claudin-1,C laud in-2, Claud in-3,C laud in-4,Cla u din-5,C laud in-7,Claud in-8 and Claudin-1 0 mRNA in mice prostate were examined by real-time fluorescence quantitative PCR. Results At 2 4 h,compared with control group,the mRNA expressions of Claudin-2,Claud in-3, Claud in-4 and Claudin-8 in combination group decreased（P〈0.05,P〈0.01）. Compared with Moschus moschiferus L. group and Frankincense group,the mRNA expressions of Claudin-2, Claud in-3, Claud in-4, Claudin-5 and Claudin-8 in combination group decreased（P〈0. 05,P〈0.01）. At 48 h,compared with control group, the mRNA expressions of Claudin-2,Claudin-3,Claudin-4,Claudin-5 and Claudin-8 in musk group decreased（P〈0. 05,P〈0.01）,the mRNA expressions of Claudin-5 in Frankincense group decreased（P〈 0. 05）,the mRNA expressions of Claudin-5 in combination group decreased（P〈 0.05）. Compared with combination group,the mRNA expressions of Claudin-2 and Claudin-8 in Moschus moschiferus L. group decreased（P〈0.05, P〈0.01）. Conclusions The combination of Moschus moschiferus L. and Frankincense can accelerate decreasing the mRNA expressions of Claudin-2, Claudin-3, Claudin-4 and Claudin-8. The Moschus moschiferus L. and Frankincense have synergetic effects.
