摘要
目的应用mat K序列对姜科10个属21种植物进行鉴定及聚类分析,为姜科药用植物的分子鉴定提供依据。方法用Snap GeneViewer校正拼接测序所得姜科的mat K序列,应用Bio Edit v7.0.9.0软件及Mega 6.0对实验所得的22条及Gen Bank数据库中下载的21条mat K序列进行比对分析,计算K2P遗传距离,构建系统进化树进行聚类分析。结果姜科的mat K序列长度约为1200bp,43条目的序列差异位点241个,不同姜科种间均存在序列差异,以美人蕉科美人蕉为外类群,构建的系统进化树显示,同属的姜科植物能聚为一类。结论利用姜科植物mat K序列数据库及遗传分析软件,所得研究结果表明mat K这一DNA条形码序列可对姜科植物进行有效地分类鉴定。
Objective In this study, we analyzed the matK sequences of 21 species among 10 Zingiberaceae genus, used to iden- tify and clustering analysis, and to provide the evidence for the molecular authentication of medicinal Zingiberaceae. Methods S- napGene Viewer was used to splice the marK sequences, BioEdit v7.0.9.0 and MEGA6.0 was used to compared and analysed the matK sequences from Genbank database and our lab, calculate the K2P genetic distance, and build system evolutionary tree. Results The lengths of matK sequences were about 1200bp, the variable sites of 43 sequences were 241, the interspecies differ- ences of matK regions were demonstrated. The system evolutionary tree built with Canna indica L. as outside group was showed that the species from the same genus are cluster for a class. Conclusion By using the database of Zingiberaceae species and ge- netic software, the marK DNA barcode sequence can be used effectively for classification and authentication of Zingiberaceae spe- cies.
