砂仁及其近缘植物化学成分的气相色谱指纹图谱研究 被引量:9

摘要 目的 建立砂仁所含挥发油的指纹图谱 ,以鉴别砂仁与常见混伪品的质量。方法 采用GC方法 ,以道地产区 10批药材为基准 ,建立砂仁挥发油的GC指纹图谱。色谱柱为HP - 5非极性柱 (30m× 0 32mm ,0 2 5 μm) ,进样口温度 2 3℃ ,检测器(FID)温度 30 0℃ ,柱温为程序升温 70℃ (停 5min) 10°C·min- 1 14 0°C(停 5 0min) ;气体流量N2 30ml·min-1,H2 30ml·min-1,空气10 0ml·min-1;流动相为N2 ,流速 0 .5 4ml·min-1;分流比为 5 0∶1。结果 建立了阳春砂挥发油成分GC指纹图谱的标准模式 ,并对常见商品混伪品进行相似度的比较 ,为建立计算机自动识别的数据库奠定了基础。结论 规范化种植基地 (GAPbase)的砂仁药材指纹图谱较为稳定 ,相似程度较高 。 OBJECTIVE To compare the fingerprints of Amomum villousm Lour with its counterfeit species collected from different areas using GC.METHODS The samples of different areas were compared by application of GC fingerprint.GC methods were optimized in order to get high quality GC profile.The GC system consisted of HP56890 and FID.The analysis was performed on an HP-5 column (crosslinked mehyl siloxame,30 mⅹ0.32 mm,0.25 μm) at the sample inlet temperature of 23℃ and FID temperature of 300℃.The temperature program of column was 70℃(5 min) to 140℃(50 min) with an increasing rate of 10℃·min -1.The carrier gas was N 2(30 ml·min -1),and the auxiliary gas were H 2(30 ml·min -1) and air (100 ml·min -1). The mobile phase was N 2 with a flow rate of 0.54 ml·min -1, and the fractional ratio of 50∶1.RESULTS After the mode of GC fingerprint was set up,comparison of similar degree to the fruits of counterfeit species has been carried on.CONCLUSION According to the selected chromatographic conditions,a good fingerprint of Amomum villousm has been established.The method is simple,accurate with good reproducibility.The samples from GAP base have high similarity,but big differences have been found between counterfeit species and standard samples.The chromatographic fingerprint of GC can be applied to control sample quality of A. Villousm.
