摘要
目的分析喉癌术后辅助放疗患者癌组织缺氧诱导因子-1α(HIF-1α)、葡萄糖转运蛋白-1(GLUT-1)表达及其与预后的相关性。方法回顾性分析,将100例喉癌术后辅助放疗患者作为研究对象,采用免疫组化法对其癌组织HIF-1α和GLUT-1表达情况进行检测。分析HIF-1α和GLUT-1表达与患者临床病理特征的相关性,并对患者进行生存分析,进而分析HIF-1α和GLUT-1表达与患者预后情况的相关性。结果喉癌组织中HIF-1α和GLUT-1阳性表达率分别为65.00%、88.00%。单因素分析结果发现,HIF-1α表达与患者临床分期、病理分级及淋巴结转移均密切相关(均P<0.05),GLUT-1表达与患者淋巴结转移密切相关(P<0.05)。多因素分析结果发现,HIF-1α和GLUT-1表达与患者预后效果密切相关(OR=6.30、1.88,95%CI:2.99~13.26、1.17~3.01,P<0.05)。Pearson相关性分析结果发现,HIF-1α和GLUT-1表达存在显著正相关关系(r=0.41,P<0.01)。结论喉癌术后辅助放疗患者癌组织HIF-1α和GLUT-1阳性表达率较高,并且两者与患者预后效果均密切相关,同时两者间的表达存在显著相关性,提示HIF-1α参与调控GLUT-1,进而在喉癌的发生和发展中起到显著的作用。
Objective To analyze the expression of hypoxia inducible factor-1 alpha(HIF-1α) and glucose transporter protein-1(GLUT-1) after adjuvant radiotherapy for laryngeal carcinoma and its correlation with prognosis.Methods 100 cases of postoperative adjuvant radiotherapy for laryngeal cancer in our hospital were selected as the research objects.The immunohistochemical method was used to detect the expression of HIF-1α and GLUT-1 in cancer tissue.The correlation between the expression of HIF-1α,GLUT-1 and the clinicopathological features was analyzed,and the survival analysis was performed,and the correlation between the expression of HIF-1α and GLUT-1 and the prognosis was analyzed.Results HIF-1α and GLUT-1 expression rates were respectively 65.00%,88.00%.Single factor analysis showed that HIF-1α expression was closely correlated with the clinical stage,pathological grade and lymph node metastasis(P < 0.05),and the expression of GLUT-1 was closely correlated with lymph node metastasis(P < 0.05).The results of multivariate analysis showed that HIF-1α and GLUT-1 expression were closely correlated with prognosis effect(OR = 6.30,1.88,95% CI:2.99 ~ 13.26,1.17 ~ 3.01,both P < 0.05).Pearson correlation analysis showed that there was a significant positive correlation between the expression of HIF-1α and GLUT-1(r = 0.41,P < 0.01).Conclusion The positive expression of HIF-1α and GLUT-1 show higher rate in patients with laryngeal carcinoma after adjuvant radiotherapy,and they are closely correlated with the prognosis,and besides the expression of HIF-1α and GLUT-1 show significant correlation,suggesting that HIF-1α is involved in the regulation of GLUT-1,which plays a significant role in the occurrence and development of laryngeal carcinoma.
