卡维地洛对血管平滑肌细胞DNA合成的影响

摘要 目的 :探讨卡维地洛对血管平滑肌细胞(VSMC)增殖的影响及其可能机制。方法 :通过贴壁细胞培养法建立豚鼠主动脉血管平滑肌细胞模型 ,应用 [3 H]TDR掺入试验研究卡维地洛对反应性氧族 (reactiveoxygenspecies ,ROS)、血管紧张肽Ⅱ(angiotensinⅡ ,AngⅡ )及胎牛血清 (fetalcalfserum ,FCS)诱导的血管平滑肌细胞增殖的影响。结果 :卡维地洛呈剂量依赖性地抑制ROS ,AngⅡ及FCS诱导的血管平滑肌细胞DNA合成 ,最大抑制率分别为 (48± 15 ) % ,(42± 7) %和 (44± 14) %。结论 :卡维地洛对ROS及AngⅡ诱导的血管平滑肌细胞DNA合成呈剂量相关性的抑制作用 。 AIM: To discuss the effect of carvedilol on DNA synthesis in vascular smooth muscle cells(VSMC) and its possible mechanism. METHODS: Rat aortic VSMC model was established in vitro,[ 3H]TDR incorporation technique was used to detect the effect of carvedilol on proliferation in VSMC induced by reactive oxygen species(ROS), angiotensin Ⅱ(Ang Ⅱ) and fetal calf serum(FCS). RESULTS: Carvedilol showed a dose dependent inhibition on DNA synthesis induced by ROS, Ang Ⅱ and FCS, the maximal inhibition rate was (48±15) %,(42±7) % and (44±14) %, respectively. CONCLUSION: The inhibition on DNA synthesis induced by ROS and Ang Ⅱ shows that carvedilol has an antiproliferative action which is probably relative to its antioxidate feature.
