摘要
为优化百蕊草离体培养快速繁殖技术,本试验以野生百蕊草的叶片及幼嫩茎段为外植体,通过研究外植体消毒时间、不同浓度激素配比对芽苗诱导、增殖、壮苗的影响,以提高外植体诱导率和瓶苗增殖率。结果表明:(1)75%的酒精溶液消毒10 s后0.1%的氯化汞溶液浸泡8 min,外植体消毒效果最佳;(2)最适初代培养配方为MS+2.0 mg·L-1 6-BA+0.13 mg·L-1 NAA,增殖继代培养配方为MS+1.5 mg·L-1 6-BA+0.3 mg·L-1 NAA,健壮无根苗培养基配方为MS+0.5 mg·L-1 NAA。
In order to optimize the rapid reproduction technology of Thesium chinense Turcz, the experiment was conducted with the leaf and young stem of wild T. chinensis as explants, and the effects of explants disinfection time and different hormones concentrations on the bud seedling induction, proliferation and growth were studied. The results showed as follows:(1) the optimal disinfection method was sterilized 10 s with 75% alcohol, and then soaked 8 min with 0.1% Hg Cl2.(2) The optimal primary culture medium formula was MS + 2.0 mg·L-16-BA + 0.13 mg·L-1 NAA, and the optimal step-generation culture medium formula was MS + 1.5 mg·L-16-BA + 0.3 mg·L-1 NAA, and the optimal robust adventitious buds culture medium formula was MS + 0.5 mg·L-1 NAA. The research could improve the value-added rate of explants induction and plantlets, providing reference for T. chinense seedling propagation.
