摘要 目的:建立高效液相-电雾式检测器(HPLC-CAD)方法测定女贞子药材中齐墩果酸和熊果酸的含量。方法:采用高效液相色谱法。色谱柱为AcclaimTM120 C18;流动相为乙腈-50mmol/L醋酸铵(70:30);流速为1.0m L/min;柱温为35℃;电雾式检测器,雾化温度35℃,采集频率10Hz。结果:齐墩果酸和熊果酸的检测质量浓度线性范围为0.3210~6.420、0.1009~2.018μg;精密度、稳定性、重复性试验的RSD<2%;加样回收率分别为100.21%~104.74%、96.26%~103.38%,RSD分别为1.78%、2.55%(n=6)。结论:该方法操作简便、稳定、重复性好,可用于女贞子中齐墩果酸和熊果酸含量的测定。 Objective To establish an HPLC - CAD method for the determination of Oleanolic Acid and Ursolic Acid in the Fruits of Ligustrum lucidum Ait. Methods HPLC was performed on the column of AcclaimTM 120C18 with mobile phase of acetonitrile - 50mmol/L Ammonium acetate solution (70 : 30) , at flow rate of lmL/min, the column temperature was 35 ℃ ; CAD : nebulization temperature was 35℃ and collection frequency was 10Hz. Results The linear range was 0. 321 0 -6. 420μg (r =0. 999 7) for Olean- olie Acid and O. 100 9 -2. 018μg (r = 0. 9999) for Ursolic Acid; RSDs of precision, stability and reproducibility tests were lower than 2.0% ; recoveries were 100.21% -104.74% (RSD = 1.78% , n =6) and 96. 26% - 103.38% (RSD =2.55% , n =6) , respectively. Conclusion The method is simple, stable and reproducible, and can be used for the content determination of Oleanolic Acid and Ursolic Acid in the Fruits of Ligustrum lucidum Air.
