鲑鱼肉和饲料中类胡萝卜素的分析方法 被引量:1

摘要 鱼肉和饲料中的虾青素分析方法对于研究鲑鱼肉色质量及饲料对其影响至关重要。本研究建立了北极红点鲑鱼肉类胡萝卜素的正相高效液相色谱分析方法。该方法采用正己烷与异丙醇等度洗脱(93∶7,体积比),流速1.1mL/min,可实现18min内4种常见鲑鱼类胡萝卜素包括虾青素、角黄素、β-胡萝卜素和玉米黄素的有效分离。研究结果显示,虾青素的回收率和变异系数值分别为97%和1.5%。正己烷和异丙醇复合溶剂提取的鱼肉样品与丙酮提取并经氨基固相萃取小柱纯化后的鱼肉样品的虾青素含量接近,但比丙酮提取样品中虾青素含量略低。正相高效液相色谱法和分光光度法测定鱼肉虾青素含量分别为1.43mg/L和2.43mg/L。研究同时建立了鲑鱼饲料虾青素分析方法。研究结果表明,-80℃冷冻结合氨基固相萃取小柱纯化可去除饲料样品中的大部分脂肪。饲料中虾青素酯皂化的最佳条件为:0.10mol/L甲基化氢氧化钠,22℃,20min;或者0.12mol/L甲基化氢氧化钠,4℃,3h。研究结果表明,两种方法可用于鲑鱼肉和饲料中的虾青素分析,为相关研究带来方便。 The carotenoind content in muscle and feed o by normal-phase and revers-phase high-performance liqu f Arctic charr (Salvelinus alpinus) was analyzed id chromatography (HPLC) as well as spectrome- tric method. Muscle sample extracted by hexane: isopropanol (HIP) or acetone was detected for astaxan- thin (AST) contents by the methods above. Results showed that a good separation of AST, canthaxanthin (CAN), β-carotene and lutein was achieved in 18 min by a normal-phase high-performance liquid chroma- tography (NHPLC)method, isocratic elution with hexane: isopropanol (HIP) (93.7, V/V) as mobile phase at a flow rate of 1.1 mL/min. Average AST recovery of 97% and AST coefficient value (CV) 1.5% were observed in fish muscle samples by NHPLC method. By the method, HiP-extraction showed similar muscular AST content (1.43 mg/L) to acetone-extracted sample cleaned with NH2-SPE column (ACEs- SPE) (1.60 mg/L), slightly lower than that in acetone-extracted sample (ACEs, 1. 81 mg/L). It was found that the NPLHC method combined with HIP extraction was used to substitute the RHPLC and ace- tone extraction for AST in salmonids muscle, which brings many conveniences to lipid measurement as well. However, AST content analyzed by NHPLC (1.43 mg/L) was lower than that by spectrophotomet- ric method (2.43 mg/L). For feed including algae, crustaceans and krill, AST appears as ester style re- quires a saponification reaction before measurement, and this usually is inhibited by the high content of lip- id in feed. The study succeeded in using deep freezing coupled with NH2-SPE column treatment for remov- al of most fat and in establishment of optimal saponifcaition condition. There was the maximal AST con- tent in 20 min saponifizion with 0.10 mol/L methic NaOH at 22 ℃ or 3 h and with 0.12 mol/L methic NaOH at 4 ℃. The findings suggest that two systematic methods involving sample extraction, fat remo- ving and NHPLC method could be used for AST analysis in salmonid muscle and feed.
