miR-218在调控肝癌发生发展中的机制及护理干预研究 被引量:2

摘要 目的分析miR-218在调控肝癌发生发展中的作用机制及护理干预对肝癌患者心理及生理的影响。方法选取2013年4月至2016年4月间于我院接受治疗的肝癌患者32例作为研究对象,并实施护理干预。另选取32例肝癌组织和相应癌旁组织为标本,以qRT-PCR法测定标本中的miR-218和Bmi-1的表达,并将miR-218前体转染HepG2细胞,以Western Blot法测定Bmi-1蛋白的表达情况。结果 miR-218在肝癌组织中的表达量显著少于癌旁组织,而Bmi-1的表达量显著高于癌旁组织(均P<0.05)。转染Hep G2细胞内Bmi-1蛋白的表达明显降低(P<0.05)。干预后患者的心理状况、疼痛状况均显著优于干预前(P<0.05)。结论肝癌细胞中miR-218表达量降低,Bmi-1表达量增加,Bmi-1为miR-218的靶向基因。护理干预可显著减少肝癌患者的心理问题,改善患者的疼痛。 Objective To analyze the mechanism of miR-218 in regulating the occurrence and development of liver cancer,and to analyze the effect of nursing intervention on the psychological and physical health of patients with liver cancer. Methods 32 cases of patients with liver cancer admitted to our hospital from April 2013 to April 2016 were selected as the research objects,and all patients received nursing intervention. Liver cancer tissues and the corresponding adjacent tissues of another 32 cases of patients with liver cancer were selected to detect the expressions of miR-218 and Bmi-1 by qRT-PCR methods; Then MiR-218 cell was transfected into HepG2 cells,and the expression of Bmi-1 protein was detected by Western Blot method. Results The expression value of miR-218 in liver cancer tissues was significantly lower than that in the adjacent tissues(P〈0.05),while the expression of Bmi-1 was significantly higher than that in the adjacent tissues(P〈0.05). The expression of Bmi-1 protein in transfected HepG2 cells reduced significantly(P〈0.05). After nursing intervention,the psychological status and pain degree of patients had significant improvement(P〈0.05). Conclusions The expression of miR-218 in liver cancer cells decreases and the expression of Bmi-1 increases. Bmi-1 is the target gene of miR-218. Nursing intervention for liver cancer patients can significantly reduce patients' psychological problems,and improve patients' pain.
