摘要
目的检测miR-218和miR-152在浆液性卵巢癌中的表达情况,并探讨其临床意义。方法取正常输卵管伞端上皮组织、良性病变卵巢组织以及浆液性卵巢癌组织,应用Real-time PCR技术检测miR-218和miR-152在上述组织中的表达情况,分析其表达与卵巢癌临床病理特征之间的关系。结果 Real-time PCR检测显示,卵巢癌组织中miR-218和miR-152的表达(分别为404.5和382.9)显著低于正常输卵管伞端上皮组织(分别为598.7和678.4)和良性病变组织(分别为523.3和587.3)(P<0.05);miR-218的低表达与卵巢癌恶性程度、肿瘤分化程度以及肿瘤转移情况相关(P<0.05),而miR-152的表达与浆液性卵巢癌各临床病理特征无显著相关性(P>0.05)。结论 miR-218和miR-152在浆液性卵巢癌组织中低表达,miR-218有可能成为浆液性卵巢癌诊断和预后的标志物。
Objective: To investigate the expression of miR-218 and miR-152 in serous ovarian cancer and its clinical significance.
Methods: The expressions of miR-218 and miR-152 in the distal epithelial tissue of fallopian tube or ovarian tissue from 50 patients with serous ovarian cancer, 20 patients with benign ovarian epithelial tumors and 10 patients with normal ovarian tissues were detected by Stem-loop real-time PCR. The correlation between the expression of miR-218 ~ miR-152 and clinieopathologic features were further analyzed. Results: The expression of miR-218 and miR-152(404.5 and 382.9)was significantly lower in serous ovarian cancer tissue than that in the distal epithelial tissue of normal fallopian tube(598.7 and 678.4)or the benign ovarian tumors tissue(523.3 and 587.3)(P〈0.05). The expression of miR-218 in serous ovarian cancer was closely related to ovarian cancer malignant degree, tumor differentiation degree and tumor metastasis status (P〈0.05). However, the expression of miR-152 was not correlated with the elinicopathologic features of serous ovarian cancer(P〉0.05). Conclusions: The expression of miR-218 and miR-152 in serous ovarian cancer is lower. The miR-218 might be potential marker for serous ovarian cancer.
