响应面法对地榆总黄酮超声提取工艺的优化

Optimization of Ultrasonic-assisted Extraction Process for Total Flavonoids from Sanguisorba Officinalis Root by Response Surface Methodology

摘要 以地榆(Sanguisorba officinalis L.)根为材料,对其所含黄酮类物质进行了提取工艺研究。通过单因素试验考察了乙醇体积分数、原料粒度、液料比、超声功率、超声时间、超声次数对地榆总黄酮提取率的影响,在单因素基础上,利用响应面法设计,优化了各项工艺参数。结果表明,超声提取地榆根总黄酮的最佳工艺条件为乙醇体积分数47.78%、原料粒度40-60目、液料比20∶1(mL∶g)、超声功率374.25 W、超声时间33.70 min。此工艺条件下,地榆根中总黄酮提取率为10.565%。按照响应面优化的最佳提取工艺条件,对40-60目地榆粉末超声提取3次,测量总黄酮提取率为11.035%。研究结果表明,超声辅助提取的黄酮得率要高于一些传统方法,适合对地榆总黄酮进行提取加工。 Using Sanguisorba officinalis root as material,the extraction technology of total flavonoids from it was studied.Through single factors experiment,the effects of ethanol volume fraction, particle size,solvent-to-solid ratio,ultrasonic power,ultrasonic time,and ultrasonic frequency on the extraction rate of total flavonoids from Sanguisorba officinalis root were investigated,and then the process parameters had been optimized by the response surface design. The results showed that the optimum extraction conditions were ethanol volume fraction of 47.78%,material particle size of 40-60 mesh,solvent-to-solid ratio of 20∶1(mL∶g), ultrasonic power of 374.25 W,ultrasonic time of 33.70 min. Under these conditions,the extraction rate of total flavonoids from Sanguisorba officinalis root was 10.565%. According to the optimum conditions,40-60 mesh powders of Sanguisorba officinalis root were extracted by ultrasonic extraction for 3 times,and the measured extraction rate of total flavonoids was 11.035%. It could be concluded that the ultrasonic-assisted extraction of flavonoids from Sanguisorba officinalis root was higher than some traditional methods,which was suitable for the extraction process of flavonoids from Sanguisorba officinalis L..
