黑麦多酚提取工艺及体外清除亚硝酸盐研究 被引量:8

Extraction Process of Rye Polyphenols and Its Scavenging Capability to Nitrite in vitro

摘要 为优化黑麦多酚提取工艺,并评价其体外清除亚硝酸盐能力,试验比较了水和乙醇对黑麦多酚得率的影响,确定乙醇为提取剂;进一步采用单因素试验和三元二次正交试验,分析乙醇浓度、乙醇用量、提取温度、提取时间对黑麦多酚得率的影响并优化工艺参数;最后对黑麦多酚体外清除亚硝酸盐能力进行测定。结果表明,体积分数50%乙醇溶液提取黑麦多酚的最优工艺参数为:乙醇用量59 mL(即料液比1:11.8),54℃水浴48 min,得率2.10 mg/g;三因素对黑麦多酚得率的主效应是乙醇用量(X_3)>提取温度(X_1)>提取时间(X_2);交互效应为X_1X_3>X_2X_3>X_1X_2;黑麦多酚具有较强的清除亚硝酸盐能力,当提取液用量超过5 mL时,清除率可达84%以上。 In order to optimize the extraction process of rye polyphenols and to evaluate scavenging capability to nitrite in vitro, ethanol was ascertained as an extracting solvent by comparing the effects of distilled water with ethanol on the extraction yield of rye polyphenols. Then, single-factor experiment and ternary quadratic orthogonal design were adopted to analyze the effects of ethanol concentration, ethanol volume, extraction temperature and extraction time on the yield of rye polyphenols and to optimize extraction process. In final, the scavenging rate of rye polyphenols to nitrite were determined. The results showed that optimum process parameters of 50% ethanol-water solution extracting rye polyphenols were as followed: ethanol volume 59 mL (i.e., the ratio of material and solvent was 1:11.8), extracted in the bath for 48 min at 54℃, the yield of rye polyphenols was 2.10 mg/g. The major effects of three factors on the yield of rye polyphenols were that ethanol volume (X3) 〉 extraction temperature (X1) 〉 extraction time (X2), the mutual effects between two factors were X1X3 〉 X2X3 〉 X1X2. Rye polyphenols had a stronger scavenging capacity to nitrite, and the scavenging rate reached above 84% when the dosage of extracting solution was more than 5 mL. The experimental results could provide theoretical foundation for deep processing of rye.
