摘要
目的:建立HPLC-PDA同时测定白术中白术内酯Ⅰ和白术内酯Ⅲ含量的方法,比较不同温度鼓风烘制与硫熏白术中这2种成分的含量,探究代替硫磺熏蒸的最佳温度鼓风烘制保存的加工方法。方法:不同温度鼓风烘制和硫磺熏蒸的白术提取物的分析采用InertsilODS-SP色谱柱,流速1.0 m L·min^(-1),流动相乙腈-水(52∶48),检测波长220 nm。结果:白术内酯Ⅰ和白术内酯Ⅲ的线性范围分别为20.4~326.4,20.9~334.4 ng(r>0.999 5),加样回收率分别为99.43%和99.20%,RSD分别为0.2%和0.3%。70℃烘制的白术中白术内酯Ⅰ和白术内酯Ⅲ的含量最高。结论:高温鼓风烘制及硫磺熏蒸均会减少白术内酯Ⅰ和白术内酯Ⅲ的含量,建议采用70℃鼓风烘制法替代硫磺熏蒸作为白术的加工方法,该方法便捷、可靠、安全,为白术的质量控制提供参考。
Objective: To establish a HPLC-PDA method for simultaneous determination of atractylenolide Ⅰ and Ⅲ in Atractylodis Macrocephalae Rhizoma,compare contents of these two constituents under sulfur-fumigation and ventilation baking,then explore a ventilation baking method to substitute sulfur-fumigation as the best processing method. Method: Analysis of Atractylodis Macrocephalae Rhizoma extract under sulfur-fumigation and ventilation baking was performed on Inertsil? ODS-SP column,mobile phase was acetonitrile-water (52:48),flow rate was 1.0 mL·min-1,column temperature was maintained at 40℃ and chromatograms were monitored at 220 nm. Result: Good linear relationships of atractylenolide Ⅰ and Ⅲ were in the ranges of 20.4-326.4 ng and 20.9-334.4 ng with r〉0.999 5,their average recoveries were 99.43% and 99.20% with RSD of 0.2% and 0.3%,respectively.Contents of atractylenolide Ⅰ and Ⅲ in Atractylodis Macrocephalae Rhizoma baked at 70℃ was the highest. Conclusion: High temperature baking and sulfur fumigation will reduce contents of atractylenolides,baking at 70℃ is the best processing method to substitute sulfur-fumigation for Atractylodis Macrocephalae Rhizoma.The method is convenient,reliable and safe.
