摘要
目的:探讨刺槐素对卵巢细胞系SKOV3细胞增殖和凋亡的影响,并分析可能的作用机制。方法:体外培养的SKOV3细胞,用不同浓度刺槐素(浓度0、4、8、16、32μmol/L)分别处理SKOV3细胞24、48、72 h,MTT法测定细胞增殖;刺槐素(浓度分别为0、8、16、32μmol/L)处理SKOV3细胞48 h后,流式细胞仪检测细胞凋亡,Western blot法检测Bax、Bcl-2、COX-2、p-NF-κB的表达。结果:刺槐素对SKOV3细胞的活性具有显著抑制作用,且呈剂量、时间依赖性;SKOV3细胞经刺槐素作用48 h后,细胞凋亡率从16.7%增加到37.5%;促凋亡蛋白Bax表达量上调,而抗凋亡蛋白Bcl-2表达量下调;刺槐素可抑制SKOV3细胞COX-2及其上游调控基因p-NF-κB的表达。结论:刺槐素对SKOV3细胞活力具有抑制作用,并可促进细胞凋亡,其作用机制可能与抑制p-NF-κB、下调COX-2表达,继而上调促凋亡蛋白Bax表达、下调抗凋亡蛋白Bcl-2表达相关。
Objective: To observe the effects of acacetin on the cell proliferation and apoptosis in ovarian cancer line SKOV3 cells and its possible mechanisms. Methods: SKOV3 cells were cultured in vitro. MTT assay and flow eytometry were used to determine the cell growth inhibitory rate and apoptosis rate of ovarian cancer cell line SKOV3 cells treated by different concentrations of acacetin at different time. Western blot were used for Bcl-2, Bax, COX-2 and p-NF- K ]3 protein expression analysis. Results: Acacetin inhibited the growth of ovarian cancer line SKOV3 cells in a dose-dependent and time-dependent manner. Acacetin could promote cell apoptosis. The apoptosis rate of SKOV3 cells increased from 16.7% to 37.5% in acacetin group. The treatment with acacetin promoted the expression of pro-apoptotic factor Bax, and suppressed the expression of anti-apoptotic factor Bcl-2. And acacetin decreased the expression of COX-2 and upstream effector of COX-2, p-NF- K B in a dose-dependent manner. Conclusion : Acacetin inhibits the cell proliferation and induces cell apoptosis in SKOV3 cells, and the effects of antitumor may be associated with inhibition of the expression of p-NF- K B, thus down-regulating the expression of COX-2, up-regulating of Bax, down-regulating of Bel-2.
