摘要
目的:研究丹参通脉方对同型半胱氨酸(HCY)诱导增殖的人主动脉血管平滑肌细胞(HA-VSMC)内质网应激(ERS)相关凋亡通路的影响,探讨丹参通脉方的心血管药理学作用机制。方法:应用HCY诱导HAVSMC增殖,使用丹参通脉方干预,Annexin V-FITC/PI法检测细胞凋亡,Western blot及实时荧光定量PCR法检测ERS通路中免疫球蛋白结合蛋白(BIP)、C/EBP同源蛋白(CHOP)蛋白及基因表达。结果:与空白组及模型组比较,丹参通脉方促进HA-VSMC凋亡;丹参通脉方可促进BIP、CHOP蛋白及基因的表达(P<0.05)。结论:丹参通脉方可促进异常增殖的HA-VSMC凋亡,其作用机制可能与促进ERS相关凋亡通路信号转导有关。
Objective: To study the effects of Danshen Tongmai Formula on HCY-induced proliferation of human aortic vascular smooth muscle cells(HA-VSMC) via endoplasmic reticulum stress(ERS) related apoptosis pathways and its mechanism of anti-atherosclerosis. Methods: The HCY was used to induce the proliferation of HA-VSMC. The Danshen Tongmai Formula was then administrated the proliferative HA-VSMC. The cell apoptosis rate was detected by Annexin V-FITC/PI method, and the amount of immunoglobulin binding protein(BIP), C/EBP homologous protein(CHOP) were quantified by Western blot and real-time reverse transcript PCR. Results: Compared with the blank and the model group, Danshen Tongmai Formula promoted the apoptosis of HA-VSMC(P<0.05). Compared with the blank and the model group, Danshen Tongmai Formula promoted the protein and gene expressions of BIP and CHOP(P<0.05). Conclusion: Danshen Tongmai Formula could promote the apoptosis of excessive proliferative HA-VSMC, which may relate to promote the transduction of ERS related apoptosis pathways.
