基于TGF-β-MAPK信号通路探讨狼疮定对SLE的治疗作用 被引量:2

Discussion on the Therapeutic Effect of Langchuangding Decoction on SLE Based on TGF-beta-MAPK Signaling Pathway

摘要 目的观察狼疮定对MRL/lpr狼疮模型鼠的转化生长因子β(TGF-β)-MAPK信号通路上的蛋白含量变化的影响。探讨狼疮定治疗SLE的内在机制,为治疗SLE提供理论依据。方法制备MRL/lpr狼疮模型鼠,取小鼠脾脏组织,提取其淋巴细胞总蛋白,再对提取总蛋白行浓度测试,然后根据蛋白浓度上样,进行蛋白免疫印迹实验,蛋白表达情况,进行结果分析。结果 Western blot法对小鼠各组脾脏淋巴细胞TGF-β2蛋白进行电泳实验结果示,正常组TGF-β2表达极少,而模型组表达明显升高,而中药组表达明显下降。正常组小鼠脾脏淋巴细胞JNK表达含量很少,而中药组表达很高,但中药组表达比模型组明显下降。正常组Erk表达很高,狼疮小鼠模型组表达下降,中药组用药后,Erk表达仍较少,与模型组区别度不明显。结论以TGF-β-MAPK信号通路为基础,根据蛋白免疫印迹法实验发现,狼疮定影响MRL/lpr狼疮模型鼠脾脏淋巴细胞总蛋白的表达,可能是狼疮定治疗SLE的机制。 Objective: To observe the effect of Langchuangding on the change of protein content in TGF-β-MAPK signaling pathway of a given MRL/lpr mouse with lupus,and investigate the inherent mechanism of treatment on SLE to provide a theoretical basis. Methods: MRL/LPR mice model with Lupus was fed and given medicine. Spleen tissues were taken and the lymphocyte of total protein was extracted,and then the extraction of total protein concentration was tested. According to the protein concentration of the sample,protein immunoblot experiments,protein expression were analyzed. Results: According to the protein immunoblot experiment,Langchuangding Decoction had a certain influence on protein expression of spleen lymphocytes in MRL/Lpr mice with lupus. Conclusion: On the basis of TGF-beta-MAPK signaling pathways,according to the protein immunoblot experiment,Langchuangding Decoction has a certain influence on protein expression of spleen lymphocytes in MRL/Lpr mice with lupus. Therefore,Langchuangding Decoction has an influence on MRL/Lpr mice with lupus,which suggests that Langchuangding Decoction has an effect on SLE.
