摘要
目的探讨层粘连蛋白5Y2（LN-5Y2）在食管癌组织中的表达及其瘤内异质性。方法采用微阵列免疫组织化学技术检测了135例食管鳞癌组织中LN-5Y2蛋白的表达情况，分析其与食管癌患者临床病理特征的关系。比较同一肿瘤不同取材位点之间、不同肿瘤同-取材位点之间LN-5Y2蛋白的表达情况。结果LN-5y2蛋白在肿瘤组织中的阳性率为40．O％（54／135），在I～Ⅱ期和Ⅲ期患者中的阳性率分别为31．1％（23／74）和50．8％（31／61），差异有统计学意义（P=0．023）。LN-5y2蛋白表达与食管鳞癌患者年龄、性别、T分期、淋巴结转移情况和分化程度无关（P〉0．05）。在54例LN．5Y2蛋白阳性的肿瘤组织中，同一肿瘤不同取材位点之间、不同肿瘤同一取材位点之间，LN．5Y2蛋白的表达均显示出一定程度的异质性。结论LN．5y2蛋白表达与食管鳞癌患者的临床分期有关．且在同一肿瘤内不同区域存在异质性。
Objective The present study aimed to evaluate the expression and intratumoral heterogeneity of LN-5Y2 in esophageal squamous cell carcinoma （ESCC）. Methods The expression of LN- 5~/2 protein was examined in 135 ESCC cases by immunohistochemistry, and to analyze its relationship with the clinical relevance of patients. The protein expressions in different regions in the same tumor as well as different nests in the same region were compared. Results Moderate and high expression of LN-5y2 protein was detected in 40.0% （54/135） of tumor tissues. Positive immunohistochemical staining was observed in 31.1% （23/74） of early stage （ stages I / Ⅱ ） cases and 50.8% （ 31/61 ） of late stage （ stageⅢ ） cases, with a significant difference between these two groups （ P = 0.023）. There was no statistical association of LN-532 expression with age, sex of patients, PT sage, lymph node metastasis and degree of tumor differentiation （P〉0.05）. However, differential expression of LN-5Y2 protein was found at different sampling sites in the same tumor and the same sampling site in different carcinomas. Conclusion High expression of LN-5Y2 is positively correlated with tumor clinical stages and there existed intratumoral heterogeneity of LN- 5Y2 expression in ESCC tissues.
